CTS-Cu Synthesis And The Effect On Growth Performance Of Weaned Piglets

This study was selected chitosan and copper sulfate as the based materials, were prepared for chitosan chelate (CTS-Cu). And the diet was fed, the effect of chitosan copper on the performance impact of weanling pig was determined, and its mechanism was discussed.A total of 120 Three-way crossbreed weanling pigs (Duroc×Landrace×Large Yorkshire), the average weight of which was about 7.8kg, were randomly divided into 4 groups with 30 piglets each according to the same parity, the same species, similar body weight, same male and female, fed with basal diet in the control group, basal diet plus 50mg/kg CTS-Cu, basal diet plus 1000mg/kg CTS-Cu and basal diet plus 200mg/kg copper sulfate in the treatments, respectively. There were 3 replicates with 10 piglets in every treatment (same male and female). Above diets were fed, respectively. The houses for pigs were undertook disinfection before the experiment. The test groups were fed by herd with the free feeding and drinking, pre-trial period of 7 days, a formal period of 30 days. The consumption and diarrhea cases were recorded per day in the test. At the start and end of the test, after 24 hours of fasting (free drinking) with weighting the pigs, then, the AFI, ADG, the feed conversion and diarrhea rate were calculated. After the feeding trial, two similar weight healthy pigs (same male and female) were randomly chosen from per repetition of each group for a total of 24 with slaughter experiment. Before the slaughter, the pigs were weight after 24 hours of fasting (free drinking). The slaughter samples based on the test need were carried out in the slaughter house. The major results showed as following:1.Preparation and Characterization of CTS-CuThe results showed that:Atomic force microscopy and infrared spectroscopy analysis showed that the chelate reaction of chitosan and Cu (Ⅱ) were occurred.2.the results of CTS-Cu in animal experiments2.1 The influence on growth performanceThe results showed that, Diets supplemented with 100mg/kg copper chitosan or 200mg/kg copper sulfate had the same effect on increasing the daily gain and lower feed conversion ratio of weaning piglets. Compared with the control group, the daily weight of groups with 50mg/kg,100mg/kg CTS-Cu or200 mg/kg copper sulfate separately increased 14.61%(P<0.05),26.28 (P<0.05,28.47% (P<0.05), and feed conversion ratios of separately lower 12.82%(P<0.05),22.05%(P<0.05)and 22.56%(P<0.05); Adding 100mg/kg copper chitosan or 200mg/kg copper sulfate can significantly increase the apparent digestibility (P<0.05) of crude protein and crude fat in feed.2.2 the impact on the copper content of inserum, tissue and fecal in pigletThe results of trace elements in serum and tissue showed that: the content of Cu in bile separately reduced by 38.72%(P<0.05) and 42.90%(P<0.05), compared groups added 50mg/kg,100mg/kg CTS-Cu with groups added 200mg/kg copper sulfate; the content of Cu in liver separately reduced 84.74%(P<0.05),77.35%(P<0.05)The determination results of trace elements in pig manure showed that:compared with the groups with 200mg/kg copper sulfate, the copper content in fecal significantly reduced in the groups with 50mg/kg,100mg/kg CTS-Cu, and separately reduced by 75.76% (4.1-fold) (P<0.05),69.41%(3.3-fold) (P<0.05).2.3 the impact on intestinal flora and intestinal morphology The intestinal flora analysis results showed that:Adding CTS-Cu and copper sulfate can significantly reduce the number (P <0.05) of E. coli, salmonella in cecum and colon, can significantly increase the amount of lactic acid bacteria (P<0.05).Light microscope observations indicated that:compared groups adding 100mg/kgCTS-Cu with that adding copper sulfate, the height of Jejunum and ileum villus significantly increased (P <0.05); the depth of crypt significantly lower (P<0.05); the height of duodenum villus and the depth of crypt has no significant differences (P>0.05).2.4 the impact on antioxidant capacity and immune functionThe determination results of antioxidant capacity showed that the T-AOC,SOD and GSH-Px of serum and liver in groups adding 100mg/kgCTS-Cu were significantly higher than that of the control group (P<0.05), while the content of MDA was decreased significantly (P<0.05). the capacity of T-AOC and GSH-Px in groups adding 100mg/kg CTS-Cu was increased by 66.30%(P<0.05,42.33% (P<0.05) compared with the groups with copper sulfate.The results of immune response showed that the index number in groups increased by 23.39%(P<0.05) for lymph node, while 9.24% (P<0.05) for C3 content in serum compared with the control group.2.5 the impact on Serum Biochemical ParametersThe results of Serum Biochemical Parameters indicated that: compared with the control groups and the groups with copper sulfate, separately, the total iron binding capacity in the groups with 100mg/kg CTS-Cu increased by 280.34%(P<0.05) and 41.98%(P<0.05) the ALP capacity increased by 20.33%(P<0.05) (in groups with 100mg/kg CTS-Cu) or 11.64 (P<0.05) (in groups with 200mg/kg copper sulfate) compared with the control group, while the groups with 100mg/kg CTS-Cu compared with 200mg/kg copper sulfate showed increasing trend, but no significant (P>0.05); the serum glucose level in groups with 200mg/kg copper sulfate increased significantly compared with the control group(P<0.05), while there was no significant between the CTS-Cu groups and the control groups (P>0.05). The ceruloplasmin concentrations in groups with 100mg/kgCTS-Cu and 200mg/kg copper sulfate were increased by 29.31%(P<0.05),12.51%(P<0.05), compared with the control group, separately.The serum hormone results showed that:the serum INS level in groups with 100mg/kg CTS-Cu increased by 30.35%(P<0.05) than that in the control groups.
