Polymorphisms In 5' Regulatory Region Of Porcine ATGL And FATP1 Gene And Their Association With Carcass And Meat Traits

Adipose triglyceride lipase (ATGL) and Fatty acid transport protein 1 (FATP1) play critical roles in the lipid metabolism process. We took ATGL and FATP1 genes as candidate factors for fat traits in pig, single nucleotide polymorphism (SNPs) in 5' regulatory region of this two genes were detected in pigs of Jinhua, Chalu black, Pietrain, Duroc, Large Yorkshire and 188 JinhuaxPietrain F2 progeny. Meanwhile, the effects of the SNPs on meat traits in JinhuaxPietrain F2 were estimated using mixed model procedure. The main results were summarized as follows:Three SNPs including A-253C→T site and two linkage mutational sites (A-845G→C and A-854T→C) were found in the-1.4 Kb of the 5'flanking region of the porcine ATGL gene by sequencing. The results of bioinformatics analysis showed that the sequence maybe include the promoter region and the A-845G→C mutation could generate or disappear the potential transcription factor binding sites, suggesting that the mutation may affect the expression of ATGL gene.The polymorphism of the A-253C→T site was detected by mispairing PCR-RFLP. The site shows the polymorphisms in pigs of Jinhua, Chalu black, Pietrain, Duroc and JinhuaxPietrain F2. The allele C had the highest frequency, but only one genotype CC was found in Large Yorkshire. Significant association was found between different genotypes with backfat thickness (BF) of the last rib, BF of gluteus medius and ham weight.The polymorphism of the A-845G→C site was detected by PCR-RFLP. Three genotypes (GG, GC and CC) were found in Chalu black, Duroc, Large Yorkshire and JinhuaxPietrain F2, while two genotypes (GG and GC) in Jinhua and only one genotype (GC) in Pietrain. Significant association was found between different genotypes with BF of shoulder, BF of the 6-7 ribs, BF of the last rib, the average BF, Loin eye area and ham weight.Three main SNPs (F-586T→C, F-1026G→A and F-1240C→A) were found in the-1.5 Kb of the 5'regulatory region of the porcine FATP1 gene by sequencing. The results of bioinformatics analysis discovered that the sequence may include the regions of promoter, CpG island, TATA box. In additionally, the F-1026G→A, F-1240C→A mutation could generate or disappear the potential transcription factor binding sites.The polymorphism of the F-586T→C site was detected by PCR-RFLP. The site show the polymorphisms in pigs of Pietrain, Duroc and Jinhua×Pietrain F2, and allele T had the highest frequency, but only one genotype TT was found in Jinhua, Chalu black and Large Yorkshire. Significant association were found between different genotypes of F-586T→C with BF of shoulder, BF of the 6-7 ribs, BF of the last rib, the average BF and leaf fat weight.The polymorphism of the F-1026G→A site was detected by PCR-RFLP. Three genotypes (AA, AG and GG) were found in Jinhua, Chalu black and Jinhua×Pietrain F2, while two genotypes (AG and GG) in Duroc, Large Yorkshire and only one genotype (AG) in Pietrain. Highly significant association were found between different genotypes of F-1026G→A with BF of the 6-7 ribs (P=0.0075).The polymorphism of the F-1240C→+A site was detected by tetra-primer ARMS-PCR. Jinhua and Jinhua×Pietrain F2 pig breeds had all genotypes while the only genotype CC in other breed. This result suggested Jinhua pig have specific characteristic. Highly significant association were found between different genotypes of F-1240C→A with intramuscular fat content (P=0.005).
