The trait of prolificy of sheep was an important approach increasing more economic benefit.In recent years,a major gene(FecB) controlling sheep prolificacy in Booroola in Australia was fond and its genetic traits,genetic efects and other characters were widely studied.During several years after this find,FecB gene was widely studied as a candidate gene in many countries in different sheep breeds.The FecB gene was found in Hu and little Han sheep in China,Which were famous prolific sheep breeds in the world.To date, Whether goat had similar gene had no clear reports.Using the method of "random sampling in typical colonies of the central area of the habitat",227 Guizhou White goats were sampled respectively from nurseryies of Dejiang, Huishui,Qinglong and Bijie contries in Guizhou Province.Using these methods of PCR-RFLP,PCR-SSCP and tetra-ARMS PCR techniques to detect the variations in the Guizhou White goats above.The aims of the present study that gene polymorphism associated with liter size and growth traits in Guizhou white goats were:1.to find the major gene for prolificacy in goat;2.to develop a method using tetra-primer ARMS-PCR for detection of Bone morphogenetic protein receptor IB(BMPR-IB) gene mutation in Guizhou white goats.The results indicate that:According to the structure analysis of BMPR-IB in human,six pairs of primer were designed to amplify from exon 5 to exon 10 taking the genomic DNA as templates extracted from 277 blood samples of Guizhou White goat.The polymorphism of the six exons in BMPR-IB gene of Guizhou White goats were detected by single strand comformational polymorphism(SSCP),restriction fragment length polymorphism(RFLP) and tetra-primer amplification refractory mutationsystem PCR(Tetra-primer ARMS-PCR). The results showed that the single nucleotide polymorphism in the coding region of glycine-serine(GS)-rich domain and serine/threonine kinase domain of BMPR-IB gene was not detected in all of the samples.The polymorphism of the six exons in BMPR-IB gene of Guizhou White goats were detected by single strand comformational polymorphism(SSCP),restriction fragment length polymorphism(RFLP) and tetra-primer amplification refractory mutationsystem PCR (Tetra-primer ARMS-PCR).The results showed that the single nucleotide polymorphism in the coding region of glycine-serine(GS)-rich domain and serine/threonine kinase domain of BMPR-IB gene was not detected in all of the samples.It suggested that BMPR-IB gene in Guizhou White goat was very conservative and those mutations found from human and sheep was rarely in Guizhou White goats.These results indicated that the conservation of BMPR-IB protein might be required for the development and differentiation of goat.
