The Polymorphism Of BMP2, BMP4, BMPR-ⅠB Genes And Their Association With Body Measurement Traits In Yellow Cattle

In order to provide theoretical foundation for beef breeding, Using PCR-SSCP and DNA sequencing, we detected the variation of the BMP2 gene, BMP4 gene and BMPR-IB gene in four Chinese indigenous cattle breeds:Qinchaun (QC, N=197), Nanyang (NY, N=85), Luxi (LX, N=104), Jiaxian Red (JXR, N=73). Association analysis of the polymorphisms with body measurement traits was done. The results showed that:1. Polymorphism in BMP2 CDS areaNo Polymorphism was detected in BMP2 CDS area.2. Polymorphism in exon 3 of BMP4 gene and its association with body measurement traitsA polymorphic microsatellite was detected in the third exon of the bovine BMP4 gene in four Chinese indigenous cattle breeds. Two genotypes (AA, AB) were identified. Clonal sequencing showed that, allele A was with 19 CA repeated dinucleotides; allele B was with 18 CA repeated dinucleotides. In the four breeds, genotype AA was the dominate genotype and its genotype frequency was more than 0.85. Least Squares Analysis revealed there were significant effects of different genotypes on withers height in the four breeds, on hip height in two breeds (LX and NY, P<0.05) and on chest circumference in QC (P< 0.05). Different genotypes did not show any significant effect on the other traits (P> 0.05). The results indicated that the microsatellite was significantly associated with some body measurement traits.3. Polymorphism in BMPR-IB gene and its association with body measurement traitsThree genotypes (AA, AB, BB) were identified in exon 8 of BMPR-IB gene in QC, LX and NY. Sequencing showed that allele A had a base mutation of T>C at the position of 43 933rd bp (based on NW-001495138), but the mutation did not cause any amino acid change. Statistical analysis indicated that in QC, LX and NY, body height of individuals with genotype BB was significantly higher than that of individuals with genotype AA and AB respectively (P<0.05). In LX and NY, waist height of individuals with genotype BB was significantly higher than those individuals with genotype AB and AA, respectively (P<0.05). In LX, body length of individuals with genotype BB was higher than those with genotype AA and AB respectively (P<0.05). In NY, body length of individuals with genotype BB was significantly higher than individuals with genotype AB (P<0.05), but there was no significant difference between BB and AA (P>0.05). There was no significant effect on the other traits (P>0.05). It concluded that BMPR-IB gene had some effects on body measurement traits and it could be used as a candidate gene for marker-assisted selection in cattle breeding.
