With the development of human life, the market demand of many kinds of food which are made from quality special-purpose wheat will rise sharply, and the HMW-GS subunit is one of the most important factors of wheat processing quality. So develop and speuial the molecular improvement of wheat quality traits has great significance for the selection of high-quality new wheat variety.In order to provide technical support for selection of high-quality new wheat variety fast and effectively, this study construct high quality and yield recurrent selection population of dwarf male-sterile wheat, transfer the new resistance and high yield gene which contain HMW-GS into the recurrent selection population of dwarf male-sterile wheat, and formulate recurrent selection technique system of dwarf male-sterile wheat which is combined with PCR molecular detection technique of HMW-GS in seedling stage. The results are as follows:1,Optimize and develop the PCR molecular detection technique system of HMW-GS subunit 5+10, 14+15 in wheat by PCR molecular detection techniqce of subunit 5+10,14+15 in wheat seedling.2,Obtain the quality recurrent selection population of dwarf male-sterile wheat with the modified HMW-GS by the methods of field planting and greenhouse and PCR molecular detection technique of subunit 1Dx5,1Bx14 in recurrent selection population of dwart male-sterile wheat hyblid and continuous backcross to subunit 5+10,14+15. Conduct PCR test on 287 plants of quality applying recurrent selection population of dwarf male-sterile wheat in seedling stage, the results show: 225 plants contain subunit 5+10, account for 78.4%; 120 plants contain subunit 14+15, account for 41.8%; 58 plants contain subunit 5+10,14+15, account for 20.2%. Conduct SDS-PAGE test on 290 plants of recurrent selection population of dwarf male-sterile wheat in grain storage protein, the results show: 227 plants contain subunit 5+10, account for 78.2%; 126 plants contain subunit 14+15, account for 43.4%; 63 plants contain subunit 5+10,14+15, account for 21.7%. The PCR molecular detection technique and the SDS-PAGE test show that this study transfer the high-quality subunit 5+10, 14+15 of HMW-GS into the recurrent selection population of dwarf male-sterile wheat successfully.3,The PCR molecular detection technique of wheat seedling HMW-GS and the SDS-PAGE test in the same poputation all show the same result. All of these results show that the PCR molecular detection technique is reliable, and the method may be an effective detection method as the molecular detection technique of wheat seedling HMW-GS.
