Isolation And Identification Of Cydia Pomonella Granulovirus Zhangye Isolate (CypoGV-zy)

This paper reported a new isolate of Cydia pomonella granulovirus (CypoGV-zy) from ZhangYe, GanSu provience in China. It's the morphological identification, molecular identification and phylogenetic analysis were reported in this thesis.1. After infection with purified CypoGV-zy, the third instar larvae of Cydia pomonella were observed by transmission electronic microscope. Granule of the CypoGV-zy was observed as elliptical shape, and the approximate measure of the granule was 400~450 nm×200 nm. It also revealed that each granule contained single rod-shaped virion with approximate size of 200~250nm×20~40nm.2. The CypoGV-zy granulin gene was obtained by PCR using the primers designed according to published CypoGV granulin genes. The granulin sequence was alignment with other reported granulin genes from different CypoGV isolates.The result showed that the sequences were the same and it was the molecular identification of the new CypoGV-zy isolate. The sequence of the CypoGV-zy granulin gene was a foundation of the following evolutionary analysis.3. The phylogenetic analysis for CypoGV-zy granulin was carried out by using the ClustalW program on the amino acid sequences of baculovirus occlusion bodies from 61 NPVs Polyhedrin and 18 GVs granulin. The results revealed that CypoGV-zy granulin has a high similarity of 85-97% with other GVs,and the similarity with NPVs was from 52% to 56%.Three species of NPVs from the genus of Neodiprion showed a low similarity with CypoGV-zy,only has a similarity of 36-37%, which suggested these three NPVs had a distant evolutionarily relationship with other baculovirus.
