Preliminary Studies On Effect Of YinChenTang For Common Carp Cyprinus Carpio

To evaluate the effect of Yinchentang(YCT) and protection mechanisms on the common carp. Four trials were designed, including the extract of YCT, the protection of YCT on common carp with the poisoning effect caused by olaquindox (feeding 42 d), effects of YCT on metabolism and health of feeding and starved carp (feeding 56 d, starved 30 d). Some indicators were detected respectively before and after starvation test, including the growth, biological character, proximate composition, serum biochemical parameter, antioxidant capacity, hepatopancrease status and histopathological observation. The results showed:(1)After rearing respectively feed of control, 0%YCT, 1.35%, 2.7% and 5.4% YCT for 42 d(all groups added 200 mg·kg-1 olaquindox except the control feed), YCT increased significantly hepatopancreas index, intestine weight ratio of carp(P<0.05); meanwhile, YCT decreased significantly serum ALT, APT activities induced by olaquindox(P<0.05), and Na +, Cl- concentrations increased significantly in YCT diet groups(P<0.05). Flow cytometry detection, YCT decreased the ratio of apoptotic cell induced by olaquindox significantly(P<0.05), and the ratio of living cell increased significantiy with the additions of YCT(P<0.05); the histopathological observation, liver cell injury was serious at 0%YCT groups compared with the control group, but the injury reduced significantly with the additions of YCT.(2) After rearing respectively feed of 0%, 0.9%, 1.35%, 4.5%YCT for 56 d, the final weight, relative growth rate, absolute growth rate at 2.7% and 4.5%YCT groups decreased significantly comparing to control group and 0.9%YCT groups, and feed efficiency increased significantly comparing to control group and 0.9%YCT groups(P<0.05); ALT levels in serum in YCT group was significantly lower than control group(P<0.05). After 30 d starvation test, the weight relief rate at 0.9%, 2.7%YCT groups decreased significantly than the control group(P<0.05), and control group were no significant differences comparing to 4.5%YCT group; change tendency of fatness degree was opposite with weight relief rate, but fatness degree at 4.5%YCT group decreased significantly(P<0.05); crude fat content of muscle at 2.7%, 4.5%YCT groups decreased significantly comparing to control and 0.9%YCT groups(P<0.05); crude fat content of hepatopancreas at 4.5%YCT group decreased significantly comparing to control group(P<0.05). Comparing between two stages, fatness degree, hepatopancreas index, intestine weight ratio, intestine body ratio, mesentery fat index, crude fat content of muscle in starvation test decreased significantly comparing to feed test(P<0.05); spleen index, ghost ratio, the content of moisture, crude protein, crude ash of muscle with all groups in starvation test increased significantly comparing to feed test(P<0.05); the content of moisture, crude protein of hepatopancreas at 0.9%, 2.7%YCT groups in starvation test increased significantly comparing to feed test(P<0.05), and at control group,4.5%YCT groups in starvation test were no significant differences comparing to feed test(P>0.05), but had a certain upward tendency; the content of crude fat of hepatopancreas at all YCT groups in starvation test decreased significantly comparing to feed test(P<0.05), but no significant differences were found between two control in two test(P>0.05); HDL, LDL, CHol in starvation test increased significantly than feeding test(P<0.05), and other index in serum decreased significantly than feeding test(P<0.05).Conclusion: YCT had a significant protection mechanisms with the carp, especially, had significant restoration and protection to liver injury induced by olaquindox. On the other hand, YCT had a certain effect in substance metabolism and energy metabolism of carp in vivo, and reduced fast consumption effect of the nutrient substance with lower dose during starvation, and promoted fatty utilization, thereby, had decreased proteinic consumption, and was conducive to the maintenance of body weight, to make weight relief rate decreased of carp after starvation test. But at higher dose, because of Rhubarb containing anthraquinones derivative, had a certain purgation effect, accordingly, caused some negative influence to fish. Meanwhile, AST, ALT level decreased after feed test, and indicated that YCT had some protection with the hepatopancreas, was a kind of very good prospects of Chinese herbal medicine preparation for the development.
