There are about 180 thousand fungus in our country, 90% have not yet been recognized, which is a very important national strategic biological resources. Polyporus pecipes, a wood rooting fungus of Polyporaceace family, is found on the trunks of living or dead trees. Pharmacologically, it is traditionally used to treat cancer such as breast cancer and prostatic cancer, and it can be eaten during the early time of its life. It has functions of enhancing immunity, softening blood vessels, preventing ageing of heart and regulating the nerve of brain. Some research results have showed that Polyporus pecipes can produce many secondary metabolites, such as alkaloid,polysaccharides,polypeptide,steroid,Triterpenoids and so on. So it is highly possible that the Polyporus pecipes would become a kind of bio-resource that have exploitation potential and prospect of application. It would play an important role in the health and agriculture area.For this purpose, used modern chromatographic techniques and isolation methods to purify the chemical constituents of ethyl acetate extract from mycelia of Polyporus pecipes systematically.In order to distill and separate the antibacterial active of Polyporus picipesand lay the foundation of application and development of biochemical pesticides, the fermentation medium of Polyporus picipes was optimized by response surface methodology which enhance the activity of antibiotics. The following are the results of the study:Eight compounds were isolated from the ethyl acetate extract of mycelia of Polyporus pecipes, of which compoundâ… was with spectroscopy identified as 1-(2,4-dihydroxy-5-methylphenyl)ethanpone, compoundâ…¡identified as ergosterol,â…¢was ergosterol peroxide andâ…¤was 4,6,8(14),22(23)-tetraen-3-one-ergostane. other compounds have not been assessed.The strongest antifungul activity of metabolites from Polyporus picipes was obtained when it was cultured in the improved liquid medium based on PDA (liquid medium based on PDA, glucose6.31g/L and yeast extract5.37g/L) with initial pH and the size of 60.74mL/250ml, Erlenmeyer flask at 150rpm and 28 for 68.32h.The results were showed that the chosen method of optimization of medium composition was efficient, relatively simple as well as time and material saving.
