Study On Characteristic Of Intramuscular Connective Tissue During Development Of Wuzhumuqin Sheep

In this study, Wuzhumuqin sheeps, from 1 to 18 month of age, were employed . The changing tendency of the content of total collagen and soluble collagen in connective tissue were investigated by methods of hydroxyproline;Changes in thermal denaturation temperature in intramuscular connective tissue were investigated by method of DSC during the development of Wuzhumuqin sheep; The changing tendency of the content of pyridinoline in intramuscular connective tissue were investigated by methods of HPLC during the development of Wuzhumuqin sheep; Proteglycans was extracted from bovine skeletal muscle through the method of CsCl density gradient centrifugation, then investigated the quantitative analysis by method of carbazole colorimetry; The main conclusions are as follow:(1)Total amount of collagen and thermal solubility of collagen in connective tissue, endomysium and perimysium were decreased during growth of sheep,the changes were significantly different(P﹤0.05). Moreover, Amount of collagen and thermal solubility of collagen in perimysium were significantly lower than that in endomysium.(2)Thermal denaturation temperature in connective tissue and endomysium were increased gradually during growth of sheep. However thermal denaturation temperature in perimysium was decreased during growth of sheep.(3)The content of pyridinoline in connective tissue increased during growth of sheep and increased slower in first 12 months ,but after the first 12months the content of pyridinoline in connective tissue is increased quicker . The changes of pyridinoline in connective tissue from different ages were significantly different(P﹤0.05).(4)The content of Uronic acid in muscle increased during growth of sheep.The changes of Uronic acid in muscle from different ages were significantly different(P﹤0.05).This study explains the propertys of connective tissue in Longissimus muscle of Wuzhumuqin sheeps during growth of sheep. Based on our study, we can provide a theoretical foundation in mechanism of changes in connective tissue Characteristics generated by the age of the sheep.
