The aquaculture industry of sturgeon and the characteristics of it's polyploidy make their interspecific hybridization feasible. But because of the lack of scientific regulatory and guidance, a large number of hybrid offspring species may cause genetic background confusion and degradation. At the same time, the disordered hybrid results in many adverse effects, such as the sturgeon breeding and species conservation.In order to promote the development of sturgeon aquaculture and international trade, it is very urgent for us to develop appropriate molecular markers for discriminating hybrid sturgeon species.Using the characteristics of maternal genetics effects and interspecific variation within species of mitochondrial DNA (mtDNA), we make comparative analysis of the 11 species of farmed sturgeon mitochondrial DNA COI gene sequence, so we can determine the female parent of hybrids of sturgeon sources. Combined with the nuclear gene simple sequence repeat (SSR) in 11 species of sturgeon, the differentiation of amplified bands provide scientific basis of the discrimination of different filial generation.1. The use of DNA barcoding technology on 11 species of farmed sturgeon.:By measuring the 11 species of sturgeon mitochondrial DNA COâ… gene sequence, and analysis of the sturgeon species and genetic distance, we constructed the phylogenetic tree. We conclude that the use of mitochondrial DNA barcoding technology not only can identify pure sturgeon species, but also can accurately determine the maternal origin of hybrids. But this method can not effectively distinguish between species of the same maternal origin.2. The use of simple sequence repeat (SSR) of nuclear genes on the 11 species of farmed sturgeon: Selecting 12 pairs of micro-satellite primers, 11 species of sturgeon simple sequence repeat were amplified, of which 6 pairs of primers can be effective among the primers in different sturgeon and all the amplified bands were different.3. Combination of DNA barcoding technology and micro-satellite DNA marker in discriminating the different species of sturgeon.:Firstly, we use the barcoding technology to classify the different species by maternal origin, and then use the micro-satellite primers to amplify each sturgeon different bands, so we can distinguish the hybrid of the same maternal and maternal sources .The results showed that, combining these two methods, the 11 species of sturgeon can be effectively discriminated.
