Citrus bacterial canker, which is serious disease, restrict the healthy development of the citrus industry. So it is important to prevent the disease. In this article, citronC05as male parent and jinlan pummelo, shatian pummelo, hongxin pummelo, bingtang sweet orange and dahong sweet orange as female parent were used in cross breeding program. Embryo rescue technology was used for embryo growth, and sequence-related amplified polymorphism (SRAP) molecular maker was used to identify their hybrids according to the polymorphism. At the same time, simple sequence repeat (SSR), Inter-simple sequence repeat(ISSR) and expressed sequence tag-simple sequence repeat(EST-SSR) and sequence-related amplified polymorphism (SRAP) were used to screening parents to get specific markers. It can provide the basis on the use of BSA with specific markers to screeen resistant and diseased pool and find related genes of citrus canker resistant. Here the following findings:(1) Jinlan pummelo,shatian pummelo,bingtang sweet orange and dahong sweet orange as female parent and citron as male parent were used in cross breeding program in2010. Stripping150fruits had got490seeds. Embryo rescue technique was used to obtain2786embryo. Then cultured629strains of seedling number and at last got the final number of survival for428strains. In2008, hongxin pummelo as female parent and citron as male parent had got24hybrids, so it finally had got452hybrids.(2) From143pairs of SRAP primers, we screened20pairs, which had specific markers of parents. Primer combinations Me6-Em5and Me10-Em12identified452hybrids, which had48real hybrids and identified12real hybrids of bingtang sweet orange,4real hybrids of dahong sweet orange,3real hybrids of jinlan pummelo,5real hybrids of shatian pummelo and24real hybrids of hongxin pummelo.(3)32pairs of SSR primers,100pairs of ISSR primers,95pairs of EST-SSR primers and143pairs of SRAP primers were used to screen specific markers of pummelo, sweet orange and citron, and had got125specific markers of citron,115pummelo specific markers and115sweet orange specific markers.
