The Studies On The ORF50 Gene Of The Spodoptera Litura Nucleopolyhedrovirus â…¡

Spodoptera litura belongs to Lepidoptera Noctuidae, It harms seri-ously to crops, including rice, maize, cotton, etc. a total of 109 families 389 kinds. Today a new Baculovirus has been found from Spodoptera litura and identified as a new Spodoptera litura baculovirus species, named litura nuclear polyhedrosis virusâ…¡(SpltNPVâ…¡). its highly reproductive rate and virulence make its killing time short ,and the Budded Virus has the fastest propagation rate in the insect body,all of these proved its characteristics and potential as a good biological pesticides.SpltNPVâ…¡has 149 Open reading frames (ORFs) in total, and inthese ORFs, ORF50 is a unique and new unknown genes, this genes has never been reported. through the NCBI BLAST online comparison, we could not find any similarity genes with it.then its encoded protein sequence was blasted online, nothing was getted neither. In the results of protein spectrum , the existence of ORF50's encoded protein gp50 was detected both in SpltNPVâ…¡ODV and BV's constituent proteins, and gp50's test score in BV is hig-her than that in ODV, associated the SpltNPVâ…¡BV's fastest infection rate in insect vivo, gp50 may play an important role in the progress of insect cells'each other infecting.pGL3-Basic was also used to analysis promoter activity of the ORF50's leader sequence 50P. The result is,with no any inducing factor, the recombinant vector pGL3-Basic-50P successfully expressed the luciferase which can produce fluorescence in the presence of fluorescent substrate.Therefore, the the leader sequence 50P was considered as a early promoter,and the ORF50 was assumed as a early gene.RT-PCR, quantitative PCR, Western blotting also be used to det-ect the gene ORF50's expression phase, it is confirmed that the transcriptions and expressions of gene ORF50 has started 2 hours later after the virus infection, Thus, we believe that the gene is an early gene. in this basis, the furtber study such as using Bacmid to obtain the corresponding deficiency of inactivation of ORF recombinant virus rescue, investigation ORF deletion, inactivation of rescue res-ponse in the infected insect virus replication system, and so on ,can be on the agenda earlier.
