This experiment was conducted to investigate the effect of Megalobrama amblycephala fed the diets containing apple seed and pumpkin seeds at three lipid levels in feed (1%, 2% and 3%)on the growth(survival rate,growth velocity,feed coefficient rate,physique and body index,Viscerosomatic index,composition of fish body),physiological function(lysozyme,LSZ;superoxide dismutase,SOD;aspartate transaminase,GOT;glutamate alanine aminotransferase,GPT;hemoglobin,Hb; Diamine oxidase,DAO;D-Lactate;Creatinine,CRE;),blood glucose and blood lipid(Triglyceride ,TG;Total cholesterol,TCHO;high density lipoprotein cholesterol,HLD-C;low-density lipoprotein cholesterol,LDL-C;glucose,GLU),study on the correlation rate(CR) betw- een fatty acid composition in tissues and organs and that of the feed.Six hundred M.amblycephala frys,weighing 14.2 g,were randomly assigned to ten dietary treatments at triplicates and each tank had twenty fish.Three dietary lipid levels(1%,2% and 3%)were tested.The control diet contained 1.5% soybean oil and the soybean oil was replaced by the inclusion of apple seed and pumpkin seeds to obtain three lipid levels.The feeding trial lasted for 60 days.On the growth,the results showed that①control with 1.5% soybean oil, used 5.1%,10.3%,15.4% apple seed in the feed,the specific growth rate decreased by 0.93%(P>0.05), 2.37% (P>0.05), 12.94% (P<0.05),feed conversion ratio increased by 0.89%(P>0.05), 2.53% (P>0.05), 18.99%(P<0.05); use 4.0%, 8.0% white pumpkin seed in the feed,the specific growth rate decreased by 1.99% (P>0.05), 8.67% (P>0.05), feed conversion ratio increased by 1.76% (P>0.05), 13.44% (P>0.05), 12.0% of white pumpkin seed in the feed,the specific growth rate increased 4.65% (P>0.05),Feed conversion ratio decreased 7.33% (P>0.05); use 2.8%, 5.5%, 8.3% of black pumpkin seed in the feed,the growth rate has increased by 2.67% (P>0.05), 0.35% (P>0.05), 0% (P>0.05), feed conversion ratio decreased by 3.64% (P>0.05), 0.35% (P>0.05), 0.66% (P>0.05).②In the protein deposition rate, the group of 15.4% apple seed decreased 18.90% than the 1.5% soybean oil group (P<0.05). In the fat deposition rate,the group of 2.8% black seed increased by 80.01% than the 1.5% soybean oil group (P<0.05).③There was no significant difference conventional ingredients(P>0.05), the groups of seeds chunky shape and fat, the hepatosomatic is smaller; On the physiological function: all the seeds groups T-SOD activity is higher than the 1.5% soybean oil group and has increased with the addition of seeds increased, the group of 8.3% black pumpkin seed T-SOD activity was significantly higher than 1.5 % soybean oil group(P<0.05); 8% white pumpkin seed group of serum GPT, GOT activity was the highest(P>0.05), while the liver GPT, GOT activity was the lowest (P>0.05); The hemoglobin of 5.1% apple seed group (79.79g ? L-1 )were significantly lower than the 2.8% black seed Group (99.11 g ? L-1 ) (P<0.05); the diamine oxidase and D lactate were no significant difference(P>0.05) ;The 4.0% white pumpkin seed group blood creatinine(10mmol / L)was significantly higher than soybean oil group(P<0.05);The blood lipid and glucose were not significantly different (P>0.05), while HDL-C have increased and LDL-C have decreased in the white and black pumpkin seeds groups; The correlation rate(CR)between fatty acid composition in tissues and organs and that of the feed showed that:①The correlation rate(CR)between fatty acid composition in organs and that in the feed grows along with the increased amount of lipid added to the feed.②There are variations in the relationship between fatty acid composition in different organs and that in the feed.The coefficients in fish body,gut fat,musle, hepatopancreas respectively were 0.72~0.88, 0.86~0.95, 0.76~0.90,0.52~0.71.③C16: 1n-7 in fish were higher than the corresponding feed; C18: 1n-9 were higher than the corresponding feed, and added the C20:1n-9, C22:1n-9; C18:2n6 was lower than the corresponding feed, C18:3n6 increased compared with the corresponding feed, and added the C20:2n6, C20:3n6, C20:4n6; C18:3n3, C20:5n3 was decreased compared to the corresponding feed, and added the C20:3n3, C22:6n3; The correlation rate(CR)between amino acid composition in fish and that of the feed showed that: in the same protein level, correlation coefficient of the amino acid composition of the fish body and muscle with the corresponding feed was no significant difference. In each group the coefficient variation of essential amino acids within 4.3%, coefficient of proline up to about 10%, and other non-essential amino acids within 4.47%These results indicated that①Apple seed, white pumpkin seed, black pumpkin seed as new lipid materials to add in feed directly is feasible, in the protein level of 28%, the appropriate quantity for the apple seed is less than 5.1%, white pumpkin seed is less than 12.0%, black pumpkin seed is less than 2.8%;②Affected by the fatty acid composition of feed, in decreasing order is gut fat, muscle, fish body, hepatopancreas. Megalobrama amblycephala can turn PUFA into SFA, MUFA, C18: 2n6 and C18: 3n3 also can be synthesized n-6 series, n-3 series high unsaturated fatty acids, but the transformation of n-3 series fatty acids is weak, while C22: 6n-3 (DHA) is more important than C20: 5n-3 (EPA) on the growth.③At the same protein level in feed, coefficient of apple seed, white pumpkin seeds, black pumpkin seeds with the fish boady and musle were not significant difference. In amino acid composition, proline is the most affected by the feed, while other amino acids affected by the feed little or not affected.
