Litchi is one of the important sub-tropical fruit trees. Embryo development status does not only determine the size of seeds and directly affect the quality of fruit and its processing quality, but also closely related to litchi production. Therefore, the study of Embryo abortion mechanisms not only to provide theoretical support to litchi breeding, and has practical value in production. This study aims at finding the information of genes associated with Embryo development by cDNA-AFLP and blotting, and offer references to the research of molecular mechanism of Litchi Embryo development and taking control measures in future.In this study ,the cDNA of three kinds of materials which include normal-type embryoes, partially abortive embryoes and fully abortive embryoes was analyzed by cDNA-AFLP. More than 60 pairs of primer combinations were Screened for constructing Embryo Development cDNA-AFLP map.Through the analysis of the map polymorphism, 60 were picked and recovered from more than 3,000 TDFs. They were sequenced, homology analyzed and blotting-verified.31 TDFs were sequenced Successfully. 7 TDFs were found to have higher homology with Embryo development-related genes through BLAST,which were TDF1(F3H), TDF2(AP4.3A), TDF3(CCR4-NOT), TDF26 ( GRP23 ), TDF38 (phospholipase gene), TDF45 (phosphatidylinositol kinase gene), TDF55 (casein kinase gene).Two TDFs were randomly selected to be verificated by blotting. The results were consistent with the electrophoresis. It is reported that these 7 genes are mainly related to signal transduction and RNA interference during Embryo development. Blotting results showed that TDF1 was detected in'Wu Ye'and'Lan Zhu',TDF38 only was detected in'Lan Zhu',which were consistent with cDNA-AFLP map. Therefore, it is speculated that regulation of signal transduction and regulation of RNA interference pathway exist in litchi Embryo development. If such two kinds of pathways are abnomal, it will lead to embryo midway-abortion.
