Broad bean is a main crop in China, which has high nutrition and medicinal value. L-DOPA is an essential drug to cure parkinson's disease. It also has some effects on curing amblyopia, fracture healing, hepatic coma, heart failure. In this study, we established the broad bean L-DOPA detection system. The effects of the factors including different varieties, parts, developmental stages, drying methods on broad bean L-DOPA contents were studied. The main results were as follows:1.Established L-DOPA detection system by HPLC method:chromatographic column:CAPCELL PAK CR,UV detection wavelength:280nm,mobile phase:acetonitrile:1‰HCOOH(5:95,V/V),flow rate:1.0mL/min,injection volume:10μL,column temperature:35℃.2.The content of L-DOPA in 197 different varieties of broad bean flowers, 52 different varieties of broad bean seeds and 32 different varieties of broad bean seedings were determined. The results showed that the content of L-DOPA in different varieties of broad bean seeds ranged from 0.009%~0.095%(8.79 mg/100g to 95.12mg/100g,DW),the highest content variety was C118,whose content was 0.095%(95.12mg/100g),2.80 times of the control ling xi yi cun(0.034%). The content of L-DOPA in different varieties of broad bean flowers ranged from 3.987% to 10.181%(DW),the highest content variety was C001,whose content was 10.181%,1.47 times of the control ling xi yi cun(16.208%). The content of L-DOPA in different varieties of broad bean seedings ranged from 4.393% to 7.640%(DW),the highest content variety was C205,whose content was 7.640%,1.38 times of the control ling xi yi cun(5.534%).3.Finding out the variation of the content of L-DOPA during the germination and seedling growth of three broad bean varieties. The content of L-DOPA decreased at first (0d) and then increased (3d,CK) during the germination. The content of L-DOPA was increased (0d) at first and then decreased (3d, CK) during the seedling growth.4.Finding out the variation of the content of L-DOPA in different parts of broad bean seedlings and flowers in different stages and different parts. The results showed that the content of L-DOPA in different parts of seedlings ranked from high to low was as follows: broad bean seedlings>root>seed. For flowers the content of L-DOPA in bud stage was higher than flowering stage and was no significant difference in flowers of different part.5.Finding out the variation of the content of L-DOPA in different drying methods of broad bean flowers. The results showed that the content of L-DOPA was no significant difference between drying at 40℃and freeze-dried. As the temperature increases, the higher temperature the lower L-DOPA content acheived. At the temperature of 100℃,the L-DOPA content (3.470%) was only 38.48%~46.77% of the control (7.419%).
