The Dmrt gene is a new gene family related to sex-determination,which palys a regulatory role in sex determination and development and found in many species. Andrias davidianus is a unique rare amphibians in China.It has important economic value and research value. Due to human factors, the resource falls sharp.Strengthen the efforts to study the hereditary substance has a great significance to preserve the Andrias davidianus.On the known sequence of Dmrt gene,the full-length cDNA sequence of Andrias davidianus Dmrt2 was obtained by using the SMART-RACE and and analyzed using bioinformatics. The full-length of Dmrt2 gene cDNA was 2 026 bp, including open reading frame of 1581 bp, a 5' untranslated region(UTR) of 58bp and a 3'-UTR of 222 bp,which can encodes 526 amino acids.Comparing the deduced amino acid sequence of Andrias davidianus Dmrt2 with 11 species in equal similarity,the result shows that that the Dmrt2 gene is more conservative. The RT-PCR result shows that the expression of Dmrt2 was observed in heart,liver,Pancreas,muscle,ovary,testis, and the expression level of Dmrt2 is the highest in the testis and heart.It points out that the Dmrt2 gene was distributed more widely,and not only involved in the regulation of sex determination,but also in maintaining other organizations, organ development and function maintenance.
