Three swine herds consisted of more than 300 sampled sows (the A herd of Large White, the B herd of Large White and the herd of Hunan Black Pig) were employed in this study. a PCR-SSCP protocol was applied to genotype the predictive T388A in sFBP gene which affirmed by an error prone PCR-RFLP, and statistic its population distribution condition for herds concerned, and analyse genotype frequencies within breeds withχ2 test. Because of the key role in the fetal development and in maternal-fetal folate transport process of the sFBP; also, bioinformatics analysis showed that the T388A resulted amino acid transformation is in a conservative region of the protein, the focus of this study is the association between the sFBP gene T388A and the litter size traits of sows, and the genetic effect. The main results are as follows:1 The gene frequencies and genotype frequencies of sFBP gene in three breedsIn the three breeds, T allele was the dominant allele, and AA is rare, in Large White pigs(B herd) found no AA homozygous sow. In Hunan Black pigs, Large White pigs(A herd) and Large White pigs(B herd), the allele frequency of A is 0.256,0.164 and 0.05. Analysis the genotype frequencies of groups withχ2 test, the results indicate that T388A is in Hardy-Weinberg equilibrium in three groups.2 The effect of sFBP gene T388A on Litter SizeThis study based on Large White(A herd), research the effect of sFBP gene on deformity, mummy and death fetus. The different genotype sows have significant difference on deformity fetus, AA genotype sows contrast with TA genotype sows and TT genotype sows, the deformity fetus are significant differences. TT and TA genotype sows are less than the AA genotype genotype sows with 0.404 and 0.397 on deformity fetus (P<0.05). On mummy and death fetus, the three genotypes was no significant difference. Analysis the effect of sFBP gene T388A on first litter size and all litter size in large white pigs(A herd), or the effect of sFBP gene T388A on all litter size in the combination group (A herd+B herd). The results show that, sFBP gene have a medium-sized additive genetic effects on TNB and NBA in Large White sows(0.3-0.4 piglets/litter), and the dominant genetic effects is significant (0.6-0.9 piglets/litter). Therefore, the three genotypes Large White sows are presented TA> TT> AA as a whole on TNB and NBA, but the difference was not statistically significant. In Hunan Black sows, research the effect of sFBP gene T388A on the performance of all litter size, the of all litter size is TT> TA> AA on TNB, on the NBA is AA> TT> TA, the above performance are no significantly differece.The results show that: sFBP gene T388A in Large White sows, on deformity fetus, A allele is significantly affected on production of deformity fetus; although, In all groups, the results obtained that the T388A of T allele is beneficial for the trend of high litter size.
