Two wheat varieties J411 and Lx987, irradiated byγ-rays and neutrons in different dosages, were used to analyse the wheat nuclear protein differential expression by 2D-DIGE to find the wheat nuclear proteins that regularted by the irradiation. Cloning and irradiation responsing expression analysis of the key gene in DNA damage repair-Ku proteins were also conducted.The main results were as following:1. There is no significant effect on the germination rate of J411 and Lx987 seeds afterγ-rays and neutron irradiation, although seedling growth was significantly inhibited in both seedling height and root length compared to the original parent controls. J411 showed more sensitivity to bothγ-rays and neutron irradiation than Lx987.2. Significant difference of wheat nuclear protein expression was revealed 85 protein spots between before and afterγ-rays irradiation. MS identification results showed proteins are involved in signal transduction, proteins synthesis,repair of photosystem and proteins associated with basic metabolism, including participation in starch synthesis , photorespiration and glycosylation. Besides, there are proteins involved in DNA replication/repair/modification and transcription /translation.3. The complete cDNA sequence of wheat Ku70/80 was cloned, as named TaKu70/80 (GQ906585 and GQ906586) for the first time. The nucleotide and amino acid sequences were analyzed. TaKu70/80 have a complete ORF respectively ,coding 626 and 706 encoding amino acid residues, with all the features of eukaryotic Ku protein domain. They also have a high homology to the Ku protein in rice and Arabidopsis.4. Real-time PCR analysis showed that the expression of TaKu70/80 are subject toγ-rays and neutron irradiation, The irradiation response patterns of TaKu70/80 may vary with different genotypes .The results also revealed some relationships between the expression and the growth time afterγ-rays irradiation.
