Protein Quality Evaluation And Innovation Of Soybean Germplasm In Zhejiang Province

Abstract/Summary:
Soybean storage protein is an important component of soybeans, which can affect nutritional quality of soybean. Vegetable soybean and processing soybean products with high protein are very popular in Zhejiang and surrounding areas. Improving soybean nutritional quality becomes one of the essential issues to be solved.In this experiment, three hundreds and forty three soybean landrance of Zhejiang province and two hundreds and sixty two soybean landrance of other provinces from the germplasm bank of HangZhou National Sub-center of Soybean Improvement were analyzed to investigate the 7S & 11S globulins contents, soybean lipoxygenase and soybean lectin, using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS PAGE), spectrophotometric and visual judging method. The main results were summarized as follows:1. Four landraces with relative lower a', a-subunit content than others were found, and some unknown small peptides between 30 KD and 17 KD appeared appeared.The results showed a great diversity in the relative contents of 11S,7S globulins and their subunits. The variation ranges of the relative content of 7S, 11S globulins and the 11S/7S ratio of soybean were 0.152-0.371,0.428-0.728 and 1.270-4.008 respectively in Zhejiang province, and the average of 11S/7S ratio is 2.291. There were five landraces 7S globulin relative content was less than 20%, while the relative content of 11S globulin higher than 60%.11S/7S ratios in seven landraces showed higher than 3.5, and protein content was higher than 45.0%. The variation ranges of the relative content of 7S, 11S globulins and the 11S/7S ratio of soybean which come from other province were 0.181-0.389,0.403-0.696,1.425-3.369 respectively, and the average of the 11S/7S ratio was 2.025, the highest and average of 11S/7S ratio of soybean landraces in the other province was lower than that of in Zhejiang province. There were twenty-six landraces in Zhejiang province and fourteen in other province that 11S/7S ratio showed higher than 3.0, and 11S/7S ratio of one landraces higher than 4.0. And the coefficient of variation in relative content of 7S and 11S/7S ratio of germplasm were greater than 0.10.The study also found that the 11S/7S ratio of soybean landraces in northern Zhejiang province and the northern part of the Huanghuai (except northeast and spring soybean of sortheast of China) were higher than that of the southern part, which indicated the distribution of the 11S/7S ratio of soybean germplasm was eco-regional. The significantly negative correlation between relative contents of 11S and 7S globulins was observed and no correlation among content of soy globulin,7S globulin 11S globulin and 11S/7S radio were found.2. The soybean lipoxygenase is beany flavor factor, we have found wild soybean germplasm ZYD04511 is Loxl and Lox2 lacking, five soybean landraces ZDD06271, ZDD06044, ZDD13834, ZDD06245 and VP35 have lower content of Lox2 or Lacking Lox2, and wild soybean germplasm ZYD04452 is Lox2 and Lox3 absence.3. The soybean lectins is one of the anti-nutritional factors. In this study,'3618'lines were radiated by 150Gy of 60Coy rays, one of radiatition'3618'lines was soybean lectins mutation and this soybean lectins mutant of seeds were stably inherited in six generation comparing normal'3618'line. We tracted soybean lectins mutation development in M6 generation from 16 days after flowering until 46 days, the lectins began synthesis at 34 days, and gradually increased until pod maturity. Lectins mutation appeared at early stage, this indicates lectins mutation may start at the molecular level.
