This study is the natural fund projects in Heilongjiang Province and Harbin City, part of the contents of scientific and technological project.The mass propagation of regeneration system for the Thymus mandschuricus Ronn'tissue culture were established by using adventitious bud and callus.The allelopathy of Thymus mandschuricus Ronn were on Pinus sylvestris var. mongolica, Larix gmelinii Rupr.,Betula platyphylla Suk.,Tagetes erecta Linn.'Yatelan'and Petunia hybrida Vilm.'Dreams'.And identified the allelochemicals which in extracting solution from Thymus mandschuricus Ronn root, stem and leaf. The main conients and results aer listed as follows:Part one:The esbatlisment of regeneration system by the tissue culture of Thymus mandschuricus Ronn.1.A regeneration protocol through stem segment has been developed for Thymus mandschuricus Ronn It had some difficulty for disinfection of explants,The results showed that the best sterilization time was 30 seconds 75% alcohol firstly,then8-10 min with merecuric chloride as antiseptics.2.The esbatlisment of regeneration system by the adventitious bud of Thymus mandschuricus Ronn.A regeneration protocol through axillary buds has been developed for Thymus mandschuricus Ronn.nodal segmengt explants of Thymus mandschuricus Ronn were cultured on Murashige and Skoog(MS) medium supplemented with different plant growth regulators (6-BA,NAA) for mass propagation.The results showed that the MS medium containing 0.5mg/L 6-BA and O.lmg/L NAA was suitable for the germination of axillary buds.The germination rate was 74% and the axillay buds grew healthily.The proliferation of axillary buds on MS medium containing 0.5mg/L and 0.01mg/L NAA was achieved to 37.6.3.The optimizing medium for pedicels explants inducing callus is MS+6-BA0.5mg/L+2, 4-D 1.Omg/L; The optimizing medium for pedicels explants inducing multiple buds induced by the route of calli is MS+6-BA0.5mg/L and GA30.1 mg/L.4.The 1/2 MS medium was suitable for the germination of radication. The best medium for rooting was 1/2 MS+IBA 0.1mg/l.The rooting ratio was 100%.5. When tissue culture plant is 2-3cm-high, the best health young plants were chosen to be toughened.Transplanted to the greenhours after hardening in climatic cabinate for 15days,the temperature condition is 26±2℃,light intensity is 3000Lux,the photoperiod is light for 14h and dark for 10h,.The survival rate reaches 76.7%.Part two Study on Allelopathy of Thymus mandschuricus Ronn.1.The infusion of haulm in concentration of 1:1 appeared inhibiting the seed germination,but in concentration of 1:15 it appeared promoting effect on Pinus sylvestris var. mongolica and Larix gmelinii Rupr.It could enhanced the seed germination rate,the effect is best when the concentration is 1:10.It appeared inhibiting the seed germination in Betula platyphylla Suk.,and Tagetes erecta Linn.'Yatelan'and Petunia hybrida Vilm.'Dreams'2.The infusion of haulm in concentration of 1:1 appeared promoting effect on the Tagetes erecta Linn.'Yatelan',both promoting effect on Betula platyphylla Suk.and Petunia hybrida Vilm.'Dreams'when in concentration of 1:10-1:15;three concentrations of infusion of root both inhibit the growth of seedlings of Pinus sylvestris var. mongolica and Larix gmelinii Rupr., promoted the growth of seedlings of Betula platyphylla Suk. and Petunia hybrida Vilm. 'Dreams',promoted the growth of root,inhibit the growth of seedling height of Tagetes erecta Linn.'Yatelan'3.The better organic extractant for the infusion of haulm and root was ethyl acetate.The main organic extracts in infusion of haulm were allelochemicals which had been studied,such as phenols,vinyl polymerization and so on.In the infusion of root,the kinds and amounts which had been find were less than in the infusion of haulm.In a word, phenols and vinyl polymerization may play a certain role in the allelopathy of Thymus mandschuricus Ronn.
