Overaccumulation of triglycerides (TG) in hepatocytes induced hepatic steatosis, the inhibition of VLDL assembly and secretion was one of reasons of the hepatic steatosis. The LXR agonist through regulate related genes to controls the VLDL assembly and secretion. In this study the primary hepatocytes of goose were used as material to study the effects of LXR agonist on intracellular and extrcellular TG level, extracellular concentration of VLDL, meanwhile, the mRNA expression of SCD1,FoxO1,MTTP,LXRα,LPL,DGAT1,DGAT2,apoB was dedected by Real-Time RT-PCR. The main results showed as follows:(1) Compared with the control group, LXR agonist at 0.01,1.0 and 10μmol/L increased intracellular TG content in a dose-dependent manner.(2) Compared with the control group, LXR agonist at 0.1,1.0 and 10μmol/L all had a significant effect on TG accumulation in goose primary hepatocytes. However, LXR agonist at 10μmol/L had an inhibiting effect on the extrcellular TG content compared with 1μmol/L(3) Compared with the control group, LXR agonist at 0.1,1.0 and 10μmol/L all had a significant effect on extracellular concentration of VLDL in goose primary hepatocytes. However, LXR agonist at 10μmol/L had an inhibiting effect on the extracellular concentration of VLDL compared with 1μmol/L.(4) In order to detect the effect of LXR agonist on assembly and secretion of VLDL-TGs, we measured the mRNA level of genes involved in the TG synthesis andassembly and secretion of VLDL-TGs in goose primary hepatocytes. compared with the control group, LXR agonist at 0.1, 1.0 and 10μmol/L had a increased effect the mRNA level of LXRα,DGAT1,LPL and SCD1; LXR agonist at 0.1,1.0 and 10μmol/L all increased the mRNA level of MTTP,DGAT2,FoxO1 and apoB genes, however, LXR agonist at 10μmol/L had an inhibiting effect on the mRNA level of MTTP,DGAT2,FoxO1 and apoB genes compared with 1μmol/L.(5)The correlation analysis showed that the intracellular TG content had a significant correlation with the gene expression of LXRα,DGAT1,LPL and SCD1 genes,the correlation coefficient was 0.990,0.998,0.985 and 0.991(P<0.01), respectively; The extrcellular TG content had a significant correlation with the gene expression of MTTP genes, the correlation coefficient was 0.983(P<0.01), and the extrcellular TG content had a correlation with the gene expression of DGAT2,apoB genes, the correlation coefficient was 0.936,0.957(P<0.05), respectively; The extracellular concentration of VLDL had a significant correlation with the gene expression of apoB,LPL genes,the correlation coefficient was 0.892,0.886(P<0.05).
