Mitochondrial antiviral signaling (MAVS) protein, localized to the mitochondrial membrane, which is down-stream of retinoic acid inducible gene-Ⅰ(RIG-Ⅰ) protein and Melanoma differentiation-associated gene 5 (MDA5) protein, can induce the expression ofβ-interferon(IFN-β) through activation of NFκB and interferon regulatory factors (IRF) to block the reproduction of virus. Thereby, MAVS protein plays a very important role in innate immunity.Broilers, Laying hens, Mountainous land chickens were housed in an isolation farm together and infected intraperitoneally on day 7 of age with 1500 plaque forming units (PFU) of the Jing-1 strain of MDV. A group of uninfected chicks were kept as negative controls in another separate farm from the infected chickens.For researching the difference of expression of MA VS gene in response to Marek's virus infaction, it was detected between tissues, such as heart, liver, spleen, lung, kidney, brain, pancreas, breast muscle, bursa of Fabricius and thymus, between any stains and groups on 4,7 and 21 days post-infection using SYBR Green real-time PCR assay respectively. The present study uncovers that:There is a difference of mortality between each strain. The mortality of Broilers is highest and laying hens is more than Mountainous land chickens. And all deaths occurred in the period of 8 DPI to 21 DPI. There is no significant difference of expression of MAVS gene between S02 and S03 strain except for between the treat groups on 7 DPI. Simultaneously with step of virus infection, the expression of MAVS gene was highly up-regulated between control group (uninfected) and treat group (infected) in Mountainous land chickens. Conversely, the expression of MAVS gene was clearly down-regulated between control group (uninfected) and treat group (infected) in Broilers. This study first detected the expression of MA VS gene in tissues in chicken, and reported that the expression of MA VS gene in pancreas is the highest and in brain is the lowest. Also, the expression of MA VS gene in bursa of fabricius and spleen is in a highly level. All of these provide strong evidences to verify the notable role of MAVS gene in innate immunity and in response to Marek's virus infection.
