Objective Although microsatellite instability (MSI) has been used to detect widely the genome of tumor human, there is no relevant reseach reports about MSI in Marek's disease virus which lead to the cancer. This thesis amis to find whether there is MSI on chromosome 4 with MDV-infected chicken embryo fibroblast (CEF) cells,and to provide basic data for the early stages of infection .Material and methods Twenty microsatellite markers on the chromosome 4 were selected. DNA were extracted from 32 CEF- samples(half of each sample infected with MDV, the other half as control ) after 5 days post- infection.With the twenty microsatellite markers the motif of DNA were amplified.All the polymerase chain reaction products were tested by 12 % denatural gel electrophoresis-silver stain.Two samples with MSI were selected for sequenceResults There were 18 microsatellite markers showing MSI in 20. the sensitivity of ABR0622 was 37.50 % , and MCW0174,ADL0266,ABR0382 were all 31.75 %,respectively; The sensitivity of ABR0622 was 37.50 % , and MCW0174,ADL0266,ABR0382 were all 31.75 %,respectively; There were 6 Microsatellite markers showing MSI higher than 20%. Six out of 20 is 30.00% . Eight out of 20(40%) markers showing MSI were found from 15% to 20%. A low incidence was found showing 3.13%, which markers were 2 out of 20 to 10%.Microsatellite instability (MSI) was found in 30 samples out of 32. The sensitivity of 11 samples were up to 45.00% ,and the sensitivity of 3 samples were up to 35.00 %, respectively. There were 16 groups showing MSI+, which were 16 out of 32 to 50 %.; There were 14 groups showing MSI-L, which were 14 out of 32 to 43.75 %.;.There were only 2 groups showing without MSI.Conclusion The microsatellite instability was found in chromosome 4 of chicken embryo fibroblast infected with MDV.There was widely MSI on the early stage of tumorigenesis with a lot of research reports. This thesis indicated the same result. The microsatellite instability in the assy maye be suggested as a valuable method for the early stages of infection with marek's disease virus ,and particularly with respect to the pathogenesis and immune control of virus-induced lymphoma in an easily accessible small-animal system.
