As the"Tree of 2008 Beijing Olympic Games", Liriodendron hybrid is a valuable tree in ornamental, which with important industrial value. The tree has a low nature maturing rate, bad reproductive, so the sprout can not meet a requirement. Somatic embryogenesis was successfully done in Liriodendron hybrids with the immature zygotic embryos as explants. As we all know, the seeds are limited for Somatic embryogenesis. Vegetative organs are available all the season, if we can establish the somatic embryogenesis with vegetative organs; the marketing supplying can be meeting.The vegetative organs were selected as initial materials, which contents biennial branch in winter, annual bud and leafs, leafstalks. Some factors had been tested about their effects on somatic embryogenesis, such as hormonal matching, Gene type, plant growth regulators culture conditions, and osmotic potential. The result showed that it is better to use 3/4MS, 2.0mg/L 2, 4-D, 0.3mg/L 6-BA to induce callus. The compact calli tissue can be obtained by two methods. One is that 0.5mg/L KT, 0.2mg/L IAA, and 0.5mg/L 6-BA to be added in basic medium; the other is with 4.0mg/L ZT, 1.0mg/L IAA. During the somatic embryogenesis, there were two elements affecting this process, ABA, light condition.Through the suspension culture of Liriodendron hybrid, the influence of shaker speeding has tested, the result showed that 95r/min was better choose. In addition, it is laying a foundation to the vegetative organ somatic embryogenesis, by the increment of callus tissue testing, and callus inducing.During in the calli inducing with vegetative organs, two genotypes, NE04 and NE55, is performed well than others. And we have more soluble protein and sugar to be found in this two. However, the true relationship between biochemistry characteristics and the somatic embryogenesis is still need to be further investigated.
