Juglans mandshurica Maxim. has beautiful ornamental, suitable for street trees and ornamental forest, it can clump planting,and also can enjoy alone, it can absorb toxic gases, improve human living environment,and it is a good landscaping tree. But as a result of J. mandshurica Maxim update is very difficult, and human disturbance,it is limited in landscape application. Using culture method is an effective way to solve the seedling endangered, obtained through somatic embryogenesis seedling can effectively expand germplasm resources. Based on J. mandshurica Maxim immature zygotic embryogenesis embryo as explant induction somatic embryogenesis, for J. mandshurica Maxim callus occurred in the process of physiological and biochemical index of difference, different concentration of carbon source, ABA, mature embryo and the influence of nitrogen source treatment on somatic embryogenesis physiological and biochemical indexes of somatic embryogenesis is studied, for establishing J. mandshurica Maxim mature embryo and plant regeneration system to provide the theory basis.1. J. mandshurica Maxim embryonic callus of soluble protein content, soluble sugar content, superoxide dismutase (SOD) and peroxidase (POD) were higher than in non embryonic callus, suggests that embryonic callus of metabolic more exuberant, organic and inorganic components from the medium, and large number of accumulation, change into energy, in case of callus differentiation.2. J. mandshurica Maxim callus of different nitrogen source are mature show that the influence of sugar higher than the different concentrations of glucose, under1.0%sugar,J. mandshurica Maxim maturity callus rate as high as72.1%, under3.0%glucose,J. mandshurica Maxim callus maturity rate reach the highest,is42.6%, it is shows that in the process of J. mandshurica Maxim embryo mature, sugar treatment is better than that of glucose.3. J. mandshurica Maxim callus of different nitrogen source are mature show that the influence of1.0g/L hydrolyzed casein processing, J. mandshurica Maxim mature callus rate was70.2%,1.0g/L glutamine treatment, callus maturity rate was62.8%, the maturation rate were similar, without obvious difference, the Juglans callus in the mature process, the two both can be replaced.4. J. mandshurica Maxim of different concentrations of ABA are mature callus showed that the influence of2.0mg/LABA processing J. mandshurica Maxim under mature callus rate up to76.1%, it grow browning, almost no differentiation ability, pale yellow, loose state.5. Different carbon source processing for the embryo in the process of mature J. mandshurica Maxim somatic embryogenesis show that sucrose treatment of soluble protein content, SOD activity, POD activity were significant difference of1.0%sucrose treatment, soluble protein content values were higher than3different concentrations of glucose disposal; Cane sugar processing under the SOD activity, POD activity was significantly higher glucose disposal, demonstrating that sucrose processing more to improve J. mandshurica Maxim embryo of resistance and adaptability.6. Different nitrogen source on the embryo in the process of mature J. mandshurica Maxim somatic embryogenesis showed that:1.0g/L casein hydrolysate under the treatment of soluble sugar content appeared the maximum of9.553mg.g-1were higher than in other different concentrations of nitrogen source;2.0g/L of casein hydrolysate and glutamine treatment under the activity of antioxidant enzyme activity down after rising first and then bimodal type change trend of rising in cotyledon and embryo period, and the highest antioxidant enzyme activity, the somatic embryogenesis of the germ resistance, adaptability is strong.7. Different concentrations of ABA treatment on the embryo in the process of mature J. mandshurica Maxim somatic embryogenesis showed that:2.0g/L ABA treatment, soluble protein content and soluble sugar content both peaked in cotyledon embryo period,2.0g/L ABA treatment, the SOD activity in the peak of heart-shaped embryo period, and a maximum of POD activity was1.0g/L ABA concentration processing, the activity is also a heart-shaped embryo period.
