Karyotype Analysis On Five Species Of Genus Buxus

Karyotype formulas of five species in genus Buxus were established and a preliminary C-banding between B. bodinieri and B. harlandii was compared in this study. At the same time, the Ag-banding of B. sinica var. parvifolia, B. bodinieri, B. harlandii and B. henryi were analyzed to verify if the satellite exists and the number of satellite. The results were as follows:1) The karyotype formula of B. sinica, B. sinica var. parvifolia, B. bodinieri, B.harlandii and B.henryi was 2n=4x=56=32m+24sm(2SAT), 2n=2x=28=20m+8sm(2SAT), 2n=2x=28=26m+2sm(SAT), 2n=2x=28=24m+4sm(2SAT), 2n=2x=28= 18m+10sm, respectively. The results showed that the five plants were mainly made up of the central centromere chromosome and near the middle centromere chromosome, which was very symmetrial and of a primitive type; B. bodinieri and B. harlandii had closer relationship, B. sinica, B. sinica var. parvifolia and B. henryi had similar origin. Moreover, results of this karyological study provided some references that could prove B. bodinieri and B. harlandii were more primitive than the other three plants.2) The results indicated that the C-banding karyotype of the B. bodinieri was 2n=28=10C + 6T~+ + 4I~+ + 2N~+ + 2CI~+I_+ + 4, the number of single set banding was 14. The C-banding karyotype of the B. harlandii was 2n=28=10C + 2T~+ + 4I~+ + 2N~+ + 2T~+I_+ + 2CI~+ + 2CT~+ + 4, the number of single set banding was 15. Generally speaking, the total numbers of the two plants was similar. The bands were mainly located in short arms and secondary constriction band was significant. But the bands of single chromosome had large differences. The results of Ag-banding karyotype indicated that there exists one satellite in chromosome from B. sinica var. parvifolia, B. bodinieri, B. harlandii and B. henry.
