To achieve comprehensive breeding objective in wheat breeding, it is required to select not only for high quality HMW-GS combinations based on no 1BL/1RS genetic background but also for ideal agronomic traits such as high yield, stress resistance and disease resistance. The aim of this study is to search for the best time and procedure of determining 1BL/1RS and HMW-GS, formulate biochemical marker technical regulation of breeding for high yield and high quality in wheat.In this study, improved A-PAGE and SDS-PAGE were used to detect 1BL/1RS and HMW-GS respectively. Nitrogen analyzer Kjeltec 2300 was applied to determine crude protein content. Settlement meter 441 was used to determine sedimentation volume according to the standard of GB/T1059-2006, and the results were all corrected to 14% flour moisture content. Glutomatic 2200 was applied to measure wet gluten content according to the standard of GB/T14608-1993. Water absorption and stabilization time were detected using farinograph 810110 according to the standard of GB/T14614-2006. The results were as follows:1. The best time of detecting HMW-GS and 1BL/1RS was found.It was first brought forward that the best times of detecting biochemical markers for 1BL/1RS and HMW-GS were 5 days and 15 days after flowering respectively. The measurement results could be applied in contemporary filed selection.2. The validity of consecutive selecting for HMW-GS in wheat was proved.Some superb HMW-GS frequency, such as 1, 5+10, 2*, 7+8, were increased by means of continual selecting, and inferior subunits was controlled.3. The quality of 5+10>2+12, 2*>1 were proved.Under the same genetic background, subunit 5+10 has better effects of sedimentation volume, absorption value and stabilization time compared with subunit 2+12. Subunit 2* has better effects of crude protein content, sedimentation volume, wet gluten, water absorption and stabilization time compared with subunit 1.4. The A-PAGE method from ISTA was modified.In order to obtain high definition electrophoretic bands, the method of A-PAGE from ISTA was partly modified. First, sample was grinded sufficiently, then extracted at 19℃-20℃for 12h. Second, separation gel density was reduced from 10.4% to 6.72%, and 115μL 0.6% H2O2 was added in 50mL gel solution.5. Biochemical marker technical regulation of selecting for high yield and high quality in wheat breeding was first brought forward.In order to select and breed high yield and high quality wheat cultivars, biochemical marker technical regulation was established. It was also brought forward about germplasm resources collecting characteristics, the principle of parent selection and the procedure of detecting HMW-GS and 1BL/1RS.
