Leaf is one of the important organs of plant. Its main function is to carry out photosynthesis. Leaf plays an important role in the form of crop yield. Therefore, studies on the mechanism of leaf development and effect of external environmental conditions on leaf development have important theoretical and practical significance. In this study, 3 materials: Longjiao951, Longjiao6 and Longjiao4 were chosen. Quantitative indexes for morphology and anatomy were set and analyzed. Mechanism of leaf development was studied on the interaction of cells. Also the effect of low temperature and low light stress on leaf morphology and anatomy quantitative indicators was studied; Changes of chlorophyll, soluble sugar, soluble protein and other physiological indexes were analyzed. We provided new ideas and methods to study the mechanism of leaf development.Under natural cultivated conditions, the capsicum leaves of three varieties, same stage and different positions, was used to study changes with the nodes increasing, and the results showed that the leaf area became larger first and then smaller; epidermal cell density and epidermal cell density / stomatal density first decreased and then increased; in unit area of leaf the cell number of palisade and spongy tissue first decreased and then increased; area of micro vein island changed first larger and then smaller; inside different types of micro vein island cell number was close to the same. The three varieties of capsicum leaves, same positions, was used to study changes with the time increasing, and the results showed that leaf area went larger; cell density in the upper and lower epidermis and cell density in the upper and lower epidermis / stomatal density decreased; in unit area of leaf cell number of palisade and spongy tissue decreased; area of micro-veins went larger; inside different types of micro vein island cell number was close to the same.In various types of micro vein island the cell number was relatively consistent, and had no change with the leaf nodes and the period varies. There were three types of micro vein island, average cell number of typeâ… was 1800 ~ 2300, average cell number of typeâ…¡was 2900 ~ 3200, average cell number of typeâ…¢was 3900~4400, from typeâ… toâ…¢cell number and area of micro vein island showed a rising trend. Conducting tissue development of micro vein island had some connection with its cell number: cell number of typeâ… was less than the critical cell number, conducting tissue inside micro vein island had not yet divided, therefore, the free end has none; cell number of typeâ…¡was close to critical cell number, and it included a free end; cell number of typeâ…¢was greater than the critical cell number, so the conducting tissue had been divided for some time, and it contained many free ends.Compared with capsicum plants under natural cultivated conditions, low temperature and low light stress would reduce the chlorophyll content, increase soluble sugar and soluble protein content. Under low temperature and low light conditions, quantitative indicators of morphology and anatomy were changed; leaf area went smaller; in unit area of leaf cell number of palisade and spongy tissue increased; area of micro vein island went smaller; inside different types of micro vein island cell number were close to the same. Size of leaf area had some connection with number and area of the micro vein island, and was independent cell number inside micro vein island, also showed that micro vein island was the basic unit of leaf.
