Change Of Cellular Immune And IL-2 MRNA Expression Inlocal Mucosal Tissues Of Probiotics Chicks Vaccinated With ND Vaccine

In this study,acid nons-pecific a-naphthyl acetate esterase staining, immunohistochemistry staining, transmission electron microscope(TEM),and real-time fluorescence quantitative PCR(SYBR GreenⅠ) were used to discover the dynamic changes of the number of TANAE+cells, CD4+T,CD8+T lymphocyte subpopulation and the expression of IL-2 mRNA in local immune tissues—gland of Harder, cecal tonsils, Peyer's patch of duodenum and ileum of probiotics chicks vaccinated with ND vaccine, which were to reveal the changes and mechanism of cellular immune in local mucosal tissues of probiotics chicks vaccinated with ND vaccine. The results were as follows:1. The numbers of T cells of gland of Harder, cecal tonsils, Peyer's patch of duodenum and ileum of probiotics chicks were enhanced compared with those of without probiotics at 4-14 days (P<0.05 or P<0.01);At 7-42 days after chicks vaccinated with ND vaccine, the numbers of T cells in above immune tissue of probiotics group, ND vaccine group and probiotics chicks vaccinated with ND vaccine were enhanced compared with blank controls group (P<0.05 or P<0.01). At the sametime, the number of T cells in ND vaccine group and probiotics chicks vaccinated with ND vaccine were increased compared with probiotics chicks (P<0.05 or P<0.01); the numbers of T cells in local immune tissue of probiotics chicks vaccinated with ND vaccine were enhanced with different degree compared with chicks alone vaccinated with ND vaccine (P<0.05 or P<0.01). The results showed that probiotics could promote the development of local immune tissue, enhanced the cellular immune function in respiratory and digestive tract mucosal, could also enhanced the immunological response to ND vaccine.2. The numbers of CD4+T cells in gland of Harder, cecal tonsils, Peyer's patch of duodenum and ileum of probiotics group, ND vaccine group and probiotics chicks vaccinated with ND vaccine were enhanced with different degree compared with blank control chicks (P<0.05 or P<0.01);the detected items above of probiotics chicks vaccinated with ND vaccine were enhanced with different degree compared with chicks alone vaccinated with ND vaccine, but no obvious difference(P>0.05); the results showed that ND vaccine vaccination enhanced the cellular immune level of local mucosal tissues. The numbers of CD8+T cells in above immune tissue of chicks vaccinated with ND vaccine were remarkably decreased compared with without ND vaccination probiotics chicks and blank control chicks (P<0.05 or P<0.01), this changes profit cellular activation, cytokine release and antibody production.3. The expression of IL-2 mRNA in gland of Harder, cecal tonsils, Peyer's patch of duodenum and ileum of probiotics group, ND vaccine group and probiotics chicks vaccinated with ND vaccine were enhanced with different degree compared with blank control chicks (P<0.05 or P<0.01); the detected items of above immune tissue of probiotics chicks vaccinated with ND vaccine were enhanced with different degree compared with chicks alone vaccinated with ND vaccine. The results showed that probiotics adding and vaccination with ND vaccine stimulate proliferation and differentiation of T cell, thus promote the expression of IL-2 mRNA.4. After challenged with ND virulent virus, all the control chicks died of the illness, morbidity and mortality rate were 100%;morbidity of probiotics chicks and ND vaccine alone vaccination chicks were 60%,20%,but both are not death; probiotics chicks vaccinated with ND vaccine have no abnormal changes. The results showed that probiotics could improve body's resistance, enhanced immune efficacy of ND vaccine makes body could better effectively resist the attack of ND virulent virus.5. Tissue ultrastructure examination revealed that the length of microvilli in cecal tonsils, duodenum and ileum of probiotics chicks, the number of mitochondria, golgi, ribosomal and rough endoplasmic reticulum and smooth endoplasmic reticulum in the cytoplasm of absorption cell were enhanced compared with those of without probiotics; the numbers of goblet cells increased, mitochondria, there are large secretory granule in acinus of gland of Hader.
