Bone morphogenetic proteins (BMP4) belonging to the transfer growth factor superfamily(TGF-β).This particular family member plays an important role in growth and development of mammals, especially for bone growth. Alternative splicing in the 5' untranslated region of this gene has been described and three variants are described, all encoding an identical protein.The aim of the present study was to assess the association of polymorphism in BMP4 gene with growth traits in three goat breeds.First,goat DNA was extracted from the blood cell in this study,three pairs of primers were designed to amplification 5' UTR,intron1,and 3' UTR region of BMP4 gene . 5'UTR Sequence Analysis indicated that a 112 nucleotide sequence conserved between goat,duckmole,human,mouse,cow and pig . some transcriptional factor binding sites including GC-box,Spl,TF,NF—KB and a CpG island located in this region. Corresponding sequence alignment of goat with cattle,pig,human and mouse demonstrated that the homologies of the nucleotides were 96.45%,90.79%,89.56%,82.26% respectively. This indicates that BMP4 is highly conservative in the species.In addition,a special palindrome sequence of 5'-CGACTAGTCG -3'was also detected.These studies provide a foundation for understanding the genetic regulation of this important gene in mammal's development. In the 3'UTR region contains a AATAAA poly-signal components, and a startup sequence of mRNA degradation ATTTA components .moreover a short sequence with more than ten continuous CA dinucleotide repeats, which was located at position157–195bp, starting at 20bp down stream from the termination site of coding region.In this study, on the basis of PCR-SSCP and DNA sequencing methods, four pairs of primers were designed to detect single nucleotide polymorphisms of 5' regulatory region, intron1,intron3 and exon 3 of BMP4 gene in both high fecundity breed (huang huai goat) and low fecundity breeds (zhong wei goat, and Boer goat) by PCR-SSCP. Two new silent mutation sites of G2203A and C2203G were identified in the intron 3, and one novel microsatellite of dinucleotide-repeated (CA) sequence was in the 3' flanking region. Six genotypes (AA, BB, CC, AB, AC and BC) were detected in the three goats.The allele BB was found to be more common in the animals investigated.Statistical analysis revealed in the 2203 and 2214 site a low polymorphism information content(PIC)in the three populations(PIC<0.25) but an intermediate PIC in the remaining three populations(0.25
