Chicken diarrhea is caused by a variety of chicken with loose stool, diarrhea characterized by the general term for a class of diseases. Poultry industry has been plagued the development of a great impact on the economic benefits, should attention. Diarrhea is caused by many and complex factors, but the dominant factors of infectious diarrhea.At the same time the disease is the common disease, and one of the most difficult prevention and control's disease. Therefore, an important pathogen causing diarrhea in chickens as soon as possible to make an accurate diagnosis, effective prevention and treatment measures as the key to controlling this disease.In this study, the starting point is by conventional microbiological methods to investigate and analysis the important pathogen of chicken diarrhea, the proportion of the bacteria, biochemical characteristics, pathogenicity, serotype and resistance, and then establishing a pathogen PCR detection methods, and clinical application, designed to provide important diagnostic chicken diarrhea condition and reliable basis.Research materials from 94 chicken farms of 20 areas in Henan Province collected 310 diarrhea materials, carried out the isolation and identification for pathogen of chicken diarrhea and resistance and PCR detection method in this subject, and achieved the expected results:1,By conventional diagnostic methods of microbiology, the 310 materials from the Henan province of 152 chickens for the major diarrheal disease pathogens isolated. Identified 124 strains of E. coli, accounts for 40%(124/310); 83 strains suspected chicken's Shigella, accounts for 26.77%(83/310); 62 strains suspected Salmonella, 20%(62/310); 41 strains of Streptococcus, accounting for 13.23%(41/310). In the separation of the four major pathogens, mixed infections was 48.02(149/310) percent ratio, the ratio of single infection 51.97%(161/310), and strains isolated from the pathogenicity test, indicating that most of the isolated pathogens are strong pathogenicity.2,On the isolated 124 strains of Escherichia coli selected 94 serological test results, the advantages serotypes of E. coli are O14, O157, O74, accounting for 57.45%(54/94) shape strain, followed by O1, O8, O9, O141, accounting for shape strain of 24.46%(23/94); 83 suspected chicken Shigella identified 18 strains of chicken's Shigella, in which dominant serotype was S.boydii and S.flexneri, accounting for 38.89% (7/18)and 33.33%(6/18); 62 suspected Salmonella strains of 14 chickens's Salmonella, of which Salmonella dominant, compared to 64.29%(9/14); 41 strains of Streptococcus chicken, D streptococcus serogroup is most, accounting for 73.17%(30/41).3,By disk diffusion method, on the separation of 14 kinds of antibiotics, pathogenic bacteria susceptibility test. The results show that the resistance of four pathogens are very serious and are mostly 10 to 13 resistance, Escherichia coli, Salmonella, Shigella chicken, chicken Streptococcus accounted for 35.11% (33/94), 41.18% ( 14/34), 45.16% (28/62), 73.17% (30/41), and serious cross-resistance phenomenon, were resistant to macrolides roxithromycin highest.In addition, E. coli mainly sulfamethoxazole / trimethoprim, cefazolin and other high resistance; Salmonella mainly quinolones and sulfamethoxazole trimethoprim resistance; Shigella bacteria mainly quinolones and minocycline resistant serious; Streptococcus mainly lincosamide and quinolone resistance is strong.4,In this study of PCR detection method, the establishment of E.coli genes by uid the PCR detection method on 124 strains of E. coli isolated tested positive rate was 90.32%(112/124); by invE build chicken Salmonella PCR gene detection methods, detection of 14 strains of Salmonella the positive rate was 100%(14/14); through the establishment of the chicken gene ipaH Shigella of PCR detection methods, detection of 18 strains of Shigella, the positive rate was 100%(18/18). The results show that the established PCR detection method is specific, sensitive and accurate and reliable, can be used to Chicken important pathogens of diarrhea detection and epidemiological investigation, has a high application value.
