Studies On Antiviral Effect And Antibacterial Effect Of Radix Isatidis Alkaloid In Vitro

We studied on the extraction technology of Radix Isatidis alkaloid, measured antibacterial and antiviral effect of Radix Isatidis alkaloid and measures antiviral effect of Radix Isatidis alkaloid by using RT-PCR;Liposoluble alkaloids and water-soluble alkaloids were extracted from Radix Isatidis with alcohol ,the content of water-soluble alkaloids was 0.044% ,and the content of Liposoluble alkaloids was 0.50%;Antibacterial effect of Radix Isatidis alkaloids was studied by cup-plate method and minimal inhibitory concentration(MIC);Their TDo,TD50,IC50 and TI to chick embryo fibrobast(CEF)cells were determined.The two kinds of alkaloids were added onto the CEF cells respective before,after, at the same time the virus or after being put together with virus for 2 hours and their in vitro antiviral effect on PPV was studied.The extraction result shows that:the content of liposoluble alkaloids and soluble alkaloids of Radix Isatidis is 0.044%and0.5%.The results shows that soluble alkaloids have little antibacterial effect , soluble alkaloids only have antibacterial effect against Salmonella and Clostridieum welchii ,while liposoluble alkaloids have good antibacterial effect against E.coli,Staphylococcu aureus,Bacillus cereus,Bacillus pyocyaneus,Clostridieum welchii.Its antibacterial diameter is 13.3±0.9mm,10.3±0.7mm,16±0.9mm,11±0.9mm,12.3±0.7mm.Radix Isatidis alkaloids have good antibacterial and antiviral effect.
