Objective(1)For measuring tissues volume fractions and catheter Feret diameters of asparagi radix to use microscopic morphometric method;(2)For measuring tissues volume fractions and catheter feret diameters of ophiopogonis radix to use microscopic morphometric method;and identify different herbs ophiopogonis radix;(3)For measuring tissues volume fractions and catheter feret diameters of isatidis radix.MethodsTransverse sections of the materials were made with polyethylene glycol 2000 as embedding media,and slicing transverse;Tissue volume fractions and catheter feret diameters of the materials were measured respectively through overlying test gridmicroscopic image on the microscopic images;Profile analysis was applied to multivariate statistical analysis of the data of the tissues volume fractions; Independent sample t-test and single factor variance analysis were applied to catheter or sclereid Feret diameters.Results(1)There were no significant differences between the volume fractions of the tissues and catheter feret diameters of three origin of the asparagi radix. So the tissues of asparagi radix had species stability. (2)There were significant differences between the volume fractions of the tissues of three origin of the ophiopogonis radix. There were no significant differences between catheter feret diameters of three origin of the ophiopogonis radix. (3)There were no significant differences between the volume fractions of the tissues and catheter feret diameters of three origin of the isatidis radix. So the tissues of asparagi radix had species stability.ConclusionThe volume fractions of the tissues can roundly response propotional relationship of asparagi radix, ophiopogonis radix and isatidis radix.So it was possible for quantitative characterization of the tissues and the catheters and sclereid of asparagi radix, ophiopogonis radix and isatidis radix. It was based to improve identification and quality of the materials.
