In this experiment, the glass horn pepper of Liling as materials, using different doses of 60Co-γirradiation processing the pepper callus, induced to grow new plants, then carry out cytological observations and molecular genetic analysis after radiation treatment. And then build a regeneration in vitro culture system which different combinations of hormones were screened to obtain the best ratio for callus in vitro studies. So as to improve the efficiency of mutation breeding, provides data reference.(1) Through the initial screening of radiation dose, set radiation treatment to three gradients:20Gy,40Gy,60Gy. After using 60Co-γinduced on the callus respectively at each dose of radiation, count out the semi-lethal dose, and take some cytological observations.(2) Using RAPD analysis on each dose of pepper DNA,82 clear bands have been amplified by 30 randomly selected primers, the average amplified band of each primer is 4.8, include 12 distinguish bands which about 14.6% of total bands. Using statistics software to analyze data, the genetic distance among each radiation gradient is from 0.934 to 0.988, the most distant two gradients are 60Gy and 20Gy, and the shortest is between 20Gy and CK.(3) In the pepper tissue culture, using cotyledon and stem as the explants respectively to induce callus. The MS based culture plus of 6-BA(3mg/L) and IAA(1mg/L) was the most suitable one for induction. The induction rate have been over 90%. For bud induce the MS+6-BA (3mg/L)+IAA (0.5mg/L)+AgNO3 (3mg/L) had the best effect, the differentiation rate have reached 75%. For root induce the MS+NAA (0.1mg/L)+IAA (0.5mg/L) was the best, differentiation rate have reached 80%. In radiation treatment on callus culture in vitro regeneration, increase the sugar content 1 time is better able to maintain cell growth, improve survival.
