The potato is a main grain and vegetable crops as well as important industrial raw materials in our country. Its economic benefits are second only to other vegetables and melon, there is widespread planting in the world. But the south early spring, often can have but actually the spring cold weather weather. This time, the South potato are entering the aerial parts fast vegetal period in idle winter field , and cause the seedling varying degree to receive the freeze injury, affecting the potato the growth. It has the obvious influence to the later period potato's yields and qualities. The potatoes create the serious freeze injury to just when the time of emergence potato because of the similarly spring cold weather in the north part of our country. In order to satisfy the production and the life demand,the cultivation is more suitable the south idle land synthesis property more superior Atlantic new variety to have the extremely vital significance. This research in view of this goal, stems of potato Atlantic were cultured in vitro. Several factors affecting stems culture and callus inducation were studied. The suitable time of ultraviolet ray was investigated. And after induced mutation or chromosomal change the material which carry on the preliminary changeability appraisal analysis.the test result is as follows:(1) The optimum medium for inducing callus from the stem's was MS+6—BA1.0mg/l+2, 4—D0.8 mg/l,in which inducing rate could reach 99.4%.(2) The optimum medium for multiplication of the stem's calluses was MS +6-BA1.0mg/L+KT0.25 mg/L+2, 4-D 0.8mg/L+NAA 0.1mg/L,in which the multiplication rate reached 5.6.(3) The optimum medium for differentiation of the stem's calluses was MS+6-BA5.0mg/L + GA3 mg/L,in which the differentiation rate could reach99.1%.(4) According to the living rate and differentiation rate of stem's calluses ,thesuitable ultraviolet radiation time was 4 min. (5) According to the living rate and differentiation rate of stem's calluses ,the suitable ultraviolet radiation and a month 4 low-temperature treatment experiment time was 3 min.Preliminary variation appraisal result: In after the processing regeneration adult plant leafs proline average content is contrast 1~1.3 times, after the processing regeneration adult plant in sugar, the chlorophyll average content is all higher than the contrast,but difference is not much remarkable. There were some plants morphologies were different from the contast. The ultraviolet ray and four low temperature processes together the plant axial bud appearing the time is earlier than the ultraviolet ray plant,and also quantity are a lot. The ultraviolet ray and four low temperature processes together the adult plant stolon the frequency is 12.4%, the ultraviolet ray processes the adult plant stolon the frequency is 43.2%, the contrast group of the frequency is 88.6%, also stolon quantity increases in turn. The ultraviolet ray and four low temperature processes together has the adult plant which the flower bud forms probably to account for the total 7.1%, ultraviolet ray processing has the adult plant which the flower bud forms probably to account for the total 4.0%, also the flower bud forms time the former is earlier than the latter. The flower bud forms early adult plant its in vivo the dissociation proline and the soluble sugar content quite is all high, the further appraisal and the examination work will wait for the consummation.This part of experiments has explored and procedure of potato Atlantic UV lure, as the foundation work has provided reference for further research in this field.
