Objective:A Pathological Model of Rats with chronic and proliferative prostatitis was established to study the effect to Saxifraga stolonifera on the model.Method:(1) Model of chronic and proliferative prostatitis:Selected male SD rats as model animal, after been anesthetized and restraint, open peritoneal cavity by sterile working. Then inject hemorrhoid injection 0.2ml to left and right lobe of prostate, while injecting equal-volume physiological saline into sham operated control group rats, and suture abdominal wall muscles and skin. Ingestation freely after 12h for operation, and sterilized wound to prevent infection. The model were determined succeed or not by compared with the normal groups, and screening the best method. (2) Physic liquor preparation:Decocting saxifrage to 100% water solution, it means there is 1g saxifrage in 1ml water solution. (3) Potency observation:Select 45 rats with similar body condition as the treatment group and divide them into 9 different groups, namely 7 days group,14 days group and 21 days group,and divide every group into high-dose group,middle-dose group and low-dose group,5 rats in every group. Meanwhile, the 15 sham operated rats for comparison were divided into 3 groups, such as 7 days group,14 days group and 21 days group,5 rats in every group. In addition, select 15 operation rats without medication as model group, and the grouping method just as the control group. (4) Intragastric administration with saxifraga stolonifera water solution in every night and morning. 1ml for a rat of low-dose group,2ml for a rat of middle-dose group and 4ml for a rat of high-dose group. (5) Determination of the treatment effect:The treatment effect could be determinated by those index as follows: pathological changes of prostate tissue, total white blood cells count and lecithin density of prostatic fluid, the activity of Serum prostatic acid phosphatase (ACP) and superoxide dismutase (SOD), and malondialdehyde (MDA) and testosterone (T) levels. Meanwhile, according to those index mentioned above, the treatment effect of different doses and sustained administration time could be compared.Results:The model of chronic prostatitis hyperplasia was successfully established by injecting hemorrhoid injection in two lobe of Prostate. Saxifrage decoction of different each dose group and sustained medication time indicated certain efficacy. Low-dose group for 7 days continue medication indicated little effect, and it could be seen that SOD and testosterone levels in blood serum were increased, ACP and MDA in blood serum were decreased, and lecithin density was rebound and inflammatory lesion of prostate was mitigated after continue medication for 14 days. As the dose increased and longer continue medication time, the treatment effect was more significant than before. The effect of high dose treatment group was most significant, especially the time of continue medication was 21 days, for all treatment indexs closing to normal,which showed great significant difference compared with the model group (P<0.01). Conclusion:Saxifrage decoction of different each dose group and sustained medication time indicated certain efficacy, as to the increased dose and extend continue medication, more significant has been discovered. Especially the time of continue medication was 21 days, for all treatment index closing to normal, which showed great significant difference compared with the model group.
