In this paper, Phalaenopsis cultivar'V31'were used to study the processes of floral bud differentiation, effect of temperature treatment on flower quality and plant growth, changes of C/N, nucleic acid and some other metabolites contents in the leaves, and mechanism of floral formation at the level of physiology and biochemistry. The main results were as follows:1) The differentiation of floral bud lasted about 60 days, which could be divided into six phases, initial differentiation, inflorescence primordium different -tiation, flower primordium differentiation, sepal primordium differentiation, petal primordium differentiation, and column and pollinia differentiation.2) Cold treatment of 30 days could accelerate early flowering, but along with the treatment time extended, early flowering were delayed, stalk lengths were shorted, floret number first increased and then decreaed. Effect of different temperature treatment on floret diameter was different, while stalk thickness between six treatments had no differences.3) The contents of soluble sugar, starch soluble protein and C/N value in the leaves decreased along with floral organs differentiation. Changes of total nucleic acid content were similar to RNA content, which declined gradually in the phase of column and pollinia differentiation, while the change of DNA content was gently.4) Based on the establishment of a set of dimensional electrophoresis system suitable for Phalaenopsis, there were 227 different spots in five protein maps of leaves at different treatment time, including 45 spots of differential expression, and nine of them were specific expression in the leaves of 30 days after treatment. There were 791 different spots in five protein maps of floral bud at different differentiation phases, including 257 spots of differential expression, fifteen of them were down-regulated expression, and 45% of them were stage-specific.
