Taking Mognolia soulangeana Soul.and M. liliflora Desr. as experimental materials, we investigated on M. liliflora pollen viability and storage conditions, self and cross pollination biology of M. soulangeana and M. liliflora, hybrid seeds germination hastening techniques, and M.soulangeana stem tip tissue culture.The results were shown as follows:1. Pollen germination and storage conditionsThe sucrose affected the pollen germination by regulating the osmotic of the medium.The plenty of pollens'wall was ruptured if there was no sugar in nutrient solution. However, pollen plasmolysis would be happened when the sucrose concentration was 15%. Actually, 5% sugar concentration was the best choice for pollen in vitro culture; B3+ affected pollen germination obviously, 1mmol L-1B3+concentration would be the best one to enhance pollen germiation; Ca2+ influenced the velocity of pollen tube growth in large content. The length of pollen tube was the longest when the Ca2+concentration was 0.01mmolL-1, and then, it would be 540μm when the pollen was cultured for 16 h.To study the pollen preservation conditions, we got the conclusion, that was, dried for 4 h , and kept 86.85% water content, the pollen would be preserved in long-term way. The pollen viability was 42.6% when it was treated in -196℃for 35d.2. The basic biology of self and cross pollination in Mognolia soulangeana Soul. and M. liliflora Desr.The Mognolia soulangeana Soul. pollen was germinated earlier than the M. liliflora Desr.'s on the stigma, and also the pollen germination rate was larger.Usually, the Mognolia soulangeana Soul. pollen was germinated after 3h pollination. But, M. liliflora pollen was germinated after 6 h pollination. The pollen tube growth velocity in M. soulangeana Soul. (♀)×M. soulangeana Soul. (♂)was the fastest among all cross combination. It arrived at the base of the ovary after 16 h pollination. However, the pollen tube of M. soulangeana Soul.arrived at the base of the M.liliflora Desr.'s ovary after 24 h pollination, and so did the M. liliflora Desr. pollen tube. The fruits-set rate in M.soulangeana Soul. (♀)×M.soulangeana Soul. (♂)was the highest, reached 90%, and the percentage of aggregate follicle with seeds was 90.5%. The fruits-set rate in M.liliflora Desr.(♀)×M.liliflora Desr. (♂)was in the next place, and the percentage of aggregate follicle and the follicle with seeds were 70%and 84.4%. The rate of fruits-set and the percentage of aggregate follicle with seeds in M.liliflora Desr.(♀)×M. soulangeana Soul. (♂) was the lowest, which could reach to 59.1% and 77.4% respectively.3. The germination conditions of hybrid seedsThe study of hybrid seeds germination conditions included seeds stratification and seeds tissue culture. In the process of seeds stratification, GA3 made the M.soulangeana Soul. (♀)×M.soulangeana Soul. (♂) seeds break dormancy quickly, and GA3+6-BA made M. liliflora Desr. (♀)×M.soulangeana Soul. (♂)seeds break dormancy quickly, but in M. liliflora Desr. (♀)×M. liliflora Desr. (♂), 6-BA was the best choice for breaking seeds'dormancy.4. Taking M.soulangeana Soul. stem tip as experimental materials for tissue culture, and that was shown, the best culture condition for M.soulangeana Soul.stem tip proliferation was MS+BA0.5mg L-1+NAA0.5mgL-1+KT1mgL-1, and the best culture condition for callus induction was MS+BA2mgL-1+2,4-D0.5mgL-1.
