Gladiolus hybridus, a type of Iris perennial flower bulbs, has long been renowned as King of Cutting Flowers all over the world. In this research, the fresh pollens of Gladiolus'Friendship'were uesd to study the best culture solution for its in-vitro culturing and germination. Based on these, it compared the advantages and disadvantages of the dyeing method, the inorganic acid method and the vitro germination method, the inorganic acid method and the vitro germination method, in order to choose appropriate method for gladiolus pollen vitality test. After the comparison and choosing of method, experiments were conducted on the effect of temperature, dehydration and way of thawing with pollens from three different cultivars( Gladiolus'Friendship',G.'Red Supreme',number 3 of Xianlin) as materials for Choosing the appropriate water content. Finally, four different cultivars (G.'Chang E Pink', G.'Rose supreme',G.'Red Supreme', G.'Amsterdam') were employed to study the determination of optimum reservation procedure. Pollens from cultivar G.'Chang E Pink'were selected to explore physiological and physiochemical changes during storage under low temperature of -80℃and 4℃. This researches lead to the conclusions as follows:(1)Dyeing method, Inorganic acid method and vitro germination method were suited to test the viability of the Gladiolus pollens. The optimum culture solution was sucrose100g/LH3BO3100mg/L+CaCl2150mg/L+Fe2(SO4)3 350mg/L+KNO350mg/L+MgSO4·7H2O100mg/L.(2)The storage temperature and water content were two important factors affecting the pollen life. The Prcedure of Gladiolus Pollen in Low- temperature storage was stated as follownig: pollen could be plunged into -80℃without drying; remove the pollen after storage time, the water at 37℃was used for Pollen thowing then.(3)During the prcess of low-temperature storage at -80℃and 4℃, peroxidation of membrane lipid was fierce in the early period. However,the contents of MDA at -80℃were lower than the contents of MDA at 4℃, which indicate the peroxidation of membrane lipid relieved slowly at -80℃; Meanwhile, the activities of SOD have no changes, while POD raised firstly and then declined; additionally, amount of soluble sugar and soluble protein increased gradually in the early period, and finally amount of soluble protein were lower than that before storage. From the whole view, all of these keep pollen vitality at some extent.
