Porcine epidemic diarrhea(porcine epidemic diarrhea, PED) is a porcine epidemic diarrhea virus (porcine epidemic diarrhea virus, PEDV) can lead to piglets caused a serious viral disease, especially in the East Asian countries is very wide spread, caused every year serious economic losses, the same as with most viral diseases prevention, control of the PED's still vaccine prevention. However, the problems currently facing is PEDV cells to adapt to difficult, complex propagation conditions, the virus titer in cell culture reproduction is low, thus a serious impediment to the PED of disease control. In this study, to improve breeding PEDV titer, increased virus yield and content of viral plaque formation, plaque formation on PEDV methods were explored, and a PEDV plaque formation method, and through the established plaque forming method PEDV LJB/03 clone strains were plaque purified, selected two cell clones of different sizes, named for the large plaque and small plaque, some biological characteristics of the virus were studied.When exploring the method of PEDV plaque formation, the plaque formation conditions are the most important, the agarose concentration is 1.5%, and the optimum temperature of the mixed cultures of culture-agarose is around 30℃, and the neutral red concentration is 0.01%, while the plaque formation conditions of the small plaque strain is different on the purification process, the small plaque formation time is longer than the large plaque, and the culture must be the newest and the stain concentration is around 0.05%.The conditions of the large plaque strain are the same as the plaque formation conditions on the process of purification.The experiments on the size of the plaque and the original virus, the virus has been studied physical and chemical properties, heat resistance test results of virus titer with increasing temperature decreases, the 4℃virus titer control was set at 37℃, 42℃, 50℃, 65℃conditions, large changes in plaque titer was significantly(P<0.01), the original conditions at 42℃virus titer of the virus changes significantly different (0.010.05); PH stability test results of large and small plaque and the original virus titers all decreased, large plaque was significantly reduced(P<0.01), the original virus and a small plaque decreased significantly(0.01
