The history of mouse genetics started when mice strains were imported from pet farm to Castle'S laboratory in Harvard University about one hundred years ago.Mouse genome is highly homologous to human and can be modified by various approaches. Moreover, thousands of inbred strains, mutant strains and implemental strains have been developed. Therefore, mouse has become the key model animal for the research of development biology, functional genomics and disease mechanism.Inbred strain of mice DDK shows a unique property of reproduction (DDK syndrome).This property is caused by an incompatibility between the cytoplasmic substance of DDK eggs and the sperm factor of other strains, both of which are controlled by the alleles at the ovum mutant (Om) locus on mouse Chromosome 11. Recently, professor Zhao found DDK strain is compatible with MOM strain (molossinus syndrome)and CASP strain (castaneus).In the meantime, the relationship between DDK strain and M.mus. musculus is not clean.In the present study, we first introduce PWK strain(musculus) to research the relationship between DDK oocytes cytoplasm and PWK sperm factors and to fulfill the relationship among four subspecies.The results as follow:1.Collecting of phenotype data:we collect 110 samples together, including 20 phenotype data of Go and 90 phenotype data of F1 backcross.These phenotype data, after compared each other, suggest that PWK male can fully fertilitize DDK females.2.Genotyping and analysis for Om locus of (N2) offspring:our study has genotyped 633 N2 samples, and calculated the (Transmission ratio of Distortion) TRD.On the basis of the form studies, the transmission ratios observed in N2 is conformed with the transmission ratios expected. Accordingly, it may be concluded that the TRD is normally when F1(DDK♀×PWK(?)) backcross DDK and B6 females.3.We confirm that the DDK oocyte cytoplasmic is compatible with PWK sperm factors after researching the phenotype data and the genotypes of Om locus of (N2) offspring.
