Objective: This study was to observe the protection of Qing NaoYi Zhi Decoction serum to the cerebral neurons when got ischemia injury. Method: Used the technology of cell culture in vitro, the fetal rat cerebral neurons were cultured. The glutamate and hypoxia injury model was made according to the mechanism of cerebral ischemia-reperfiision injury. With the method of In Situ Cell Death Detection (TUNEL), we examined the influence of Qing NaoYi Zhi Decoction serum to the cell apoptosis when glutamate injury. IL-6 gene was measured with the method of Reverse Transcription -Polymerase Chain Reaction (Rt-PCR) when got hypoxia and glutamate injury. We also observed the influence of the herb and Taurine serum to IL-6. The image software analyzed the results. The cell structure were detected through phase contrast microscope (PCM) and scanning electron (SEM) Result: 200 ii M glutamate cultured with neuron for 10 mm can represent the excitotoxicity of glutamate. The apoptosis rate induced by this method is most obviously after reculture for 3-6 hours. Qing NaoYi Zhi Decoction serum can manifestly reduced the ratio of glutamate induced neuron apoptosis (p<0.05). Glutamate group is 47.28% and the Chinese matria medica group is 24.76%. It can regulate 11-6 gene expression to the normal immunology level when got injury. The prevention group has better effect than the cure group. The decoction shows more effective than Taurine in these fields. Conclusion: From the glutamate injury model dynamic observing, point out the therapeutic time window of cerebral ischemia should be within 6 hours. Qing NaoYi Zhi Decoction can protect culture fetal rat neurons against glutamate and hypoxia induced apoptosis through the regulating of interleukin-6 gene expression. It has a significant protective effect on the cerebral neurons. Proved the important role of prophylatic neuroprotection to stop the ischemic cascades. This research has provided experimental evidence to the prevention and cure to the wind stroke and the mechanism of vascular dementia's occur and develop.
