Research Of The Effect Of PQS On The Expression Of Apoptosis-Related Gene P53 And XIAP On Cerebral Ischemia Of Rats

Objective: To study and explore the neuroprotective effects and the further mechanism of Panax quinquefolium saponins from steams and leaves(PQS)on cerebral ischemia in rats.Methods: The male rats were pretreated with PQS 50 and 100㎎·㎏ ^-1 ig, one time per day for 7d, and then subjected to focal cerebral ischemia damage induced with fishing lines in the middle cerebral artery ( MCA). In order to observe the neuroprotective effect of PQS on cerebral ischemia,at 24h after cerebral ischemia ,the neurological deficit were deterinimed by Longa's score; HE staining was used to observe the changes of histomorphology; the effects of PQS on protein expression of p⁵³ and XIAP was detected with enzyme-linked immuno sorbent assay ( ELISA ) method;In addition, flow cytometry were used for determination of the cortical neuronal apoptosis;we observed the gene expression of p⁵³ mRNA and XIAP mRNA with reverse transcription polymerase chain reaction (RT-PCR).Results: PQS could make obviously pathomorphology of cerebral ischemia better, improve change of ethology, decrease the rate of cell apoptosis of ischemic cerebral tissue, inhibit the expression of p⁵³ mRNA and protein and increase the expression of XIAP mRNA and protein in cerebral cortex after cerebral ischemic injury of rats. Conclusion: PQS has protective effects on focal cerebral ischemia damage and the mechanism of PQS against cerebral ischemia is related to inhibiting cell apoptosis and affecting the related expression of gene and protein interrelated with cell apoptosis.Key Words: Cerebral Ischemia;Panax quinquefolium saponins from steams and leaves ( PQS); Cell Apoptosis; Neuroprotective Effect;...
