Objective: Recently, more and more attention has been focused on the significance of antiapoptotic mechanism under multidrug resistance (MDR), because people are now getting fresh knowledge of the mechanism how tumor cells acquire drug resistance, thanks to our in-depth studies of antiapoptotic gene cl-2. Actually, there are many more chemotherapeutic drugs having anticancer effect by way of inducing the apoptosis of tumor cells. However, the rising-up of the expression of Bcl-2 gene does obviously enhancing existance of the tumor cell line against such inducing, which displays a close connection between the enhancement of expression and the formation of MDR, put in other words, such enhancement really plays such a magnificant role in tumor cell drug resistance mechanism. Unfortunately, it still remains unclear to us what the mechanism concerning renal cancer primary resistance against chemotherapeutic drugs. Henceforth, the investigation of human renal cancer drug resistance mechanism enjoys value both in theory and clinic for our understanding and cure of renal cancer. Therefore, this experiment was designed to make a probe, starting from the apoptosis, into renal cancer occurrence mechanism of multidrug resistance, i.e. in Bcl-2 gene role in multidrug resistance of human renal cell carcinoma cell line. Methods: Through a cell culture in vitro, different concentration ADM (ranging from 0.1, 1.0 to 1 O.Omg/L) are produced to induce the apoptosis of human renal cell carcinoma cell lines, one being the high drug resistance to ADM(GRC- 1), the other the low drug resistance to ADM(RCC-925). And then, both the flow cytometry and the immunocytochemistry are adopted to make researches into apoptosis by ADM- induced GRC-l & RCC-925, and into the expression of Bcl-2 gene. Finally, observeto find out whether or not there exists sufficient distinction in terms of the two cell lines sensitivity towards ADM-induced apoptosis ,and whether or not such distinction is related to the expression of Bcl-2 gene in tumor cell lines. Results: (1) Under the one and same circumstance, compared with that of RCC-925 cells, apoptosis of GRC-1 cells is apparently inhibited. (2) Regarding the expression of Bcl-2 gene, GRC- 1 behaves stronger than RCC-925. Conclusions: It is the high expression of Bcl-2 gene in multidrug resistance of human renal cell carcinoma cell line that constitutes the major cause rendering apoptosis inhibited, which fortells the significance for the creation of multidrug resistance of human renal cell carcinoma cell line.
