OBJECTIVE: Multidrug resistance/*/ (MDR) is considered to be the major obstacle for chemotherapy since people fond it half century ago. In order to reverse tumor MDR in vitro, we designed this study to establish human multidrug-resistnat bile duct carcinoma cell line (FRH-0201/Adm) and to investigate its biological characteristics, making a base for the further study of the mechanism.of the MDR and the reverse of the MDR.METHODS: An adriamycin-resistant bile duct carcinoma cell subline (FRH-0201/Adm) established in vitro using intermittently 2 μ g/ml concentration of adramycin in culture, cell growth was measured and multidrug resistance to multianticancer agents was evaluated by MTT assay. Flow cytometry(FCM) was performed to determine cell cycle, and to assess the expression of p-glycoprotein.through immunohistochemistry(IHC).MDR mRNA expression related to this protein was determined with transcription polymerase chain reaction(RT-PCR). analysis of the cell cycle by Flow cytometry (FCM), assays of tumor marker using enzyme-linked immunosorbent assay (ELISA), the fluorescence density of adramycin.analysed by laser scanning confocal microscope(LSCM)...
