Glioma is the most frequen tumor in the central nervoussystem .The genefating and the development of glioma areconsidered to be a 1oss contrl of growth of the cell with multiplefacors and multiple phases. Proto oncogene and suppress oncogeneplay a bootent role in the contrlling cell's growth. HSP70 is aspecial protein produced in oguism when it's circumstriceexacerbated, it has biological actiVities and multiple physiologicalfunctions. It can protect organism (or cell) from being deStroyed orto be slightiy destroyed. There are almost no HSP70asA orHSP70 being detected in normal bran, bat in malignat boors,HSP70 are hghly expressed. HSP70 and it's protein products canregulate the prolifefatiqn of cell by the inteedion with Protooncogenes, suppress oncogenes tal their protoin Products, it alsoplays a impotent role in the generation and evolving of malignantumors. The levels of ploidy in cells can reflect the proliferation oftumors cell, therefore, the main method which is the mostfrequently used is to test the DNA ploidy for inferring theproliferating condition of tumors, it includes DNA index,Aneuploid rate etc. These metfical information are very objectiveand can reduce the errors caused by the mistake of subjectivejudgment in the pathological working to a certain extent. Thepurpose of our stlldy is through the research of the correlation andthe inherent links among the expression of HSP70, DNA ploidy andthe pathological grading of glioma by comParing them one withanother,not only to improve the accuracy of the diagnosis of glioma,but also to give use to prognosis and the therapy of glioma.40 brain glioma patieflts from 1992 to 1999 were collected inour study. All of them had been pathologically diagnosed. Pick upthe glioma paraffin specimens, slice a piece of specimen iflto 4um atthickness. Use the immunohistochemical method to test the.expression of HSP70 in these 40 specimen. The results shew thatthe positive rate in the group of high grade glioma (80%) wasapparently higher than that in low grade glioma (45%) and theDNA ploidy in group of HSP70 highly eXPressed (+++) was alsoapparently higher than that in grouP of lowly expressed. Theircorrelation were positive. So we could infer that the accumulation ofHSP70 in tumor cells is one of signa1s which reflect the evolvingof malignancy of g1ioma.Further study of the significance of HSP70 will be useful tothe gene theraPy ofglioma. From the same 40 specimens,we made tWo sets of slices,one at 4um of thickness, another at 8um of thickness. After Feulgenstaining the DNA tuthed into purp1ish red. We used the true colorpathological micro-image analyzing system produced bymicroelectric technique research institllte, Beijing, to test the DNAploidy (DI, 5cER). The result shew that the average of DI, 5cERin high grade glioma were higher than that in low grade glioma,there was significant difference bforeen the tWo averages. Thepositive rate of 5cER between two grouP was also significamlydifferent, the correlation was positive betWeen DNA ploidy and thegrading ofglioma. So we could infer that the change of DNA ploidyis a impotent signal which indicate the evolving of malignancy ofglioma, it can reflect understand glioma me biological behavior ofglioma, helping us to more correctly.
