Basic fibroblast growth factor, a member of neurotrophic factors, has been noticed for its role in central nervous system injury. The aim of study is to investigate pattern of bFGF in spinal cord injury and its role in nitric oxide signal system, so as to provide scientific evidence for its clinical application.In the present study, we observed the level of basic fibroblast growth factor (bFGF) gene expression and the effect of exogenous bFGF on nitric oxide levels, nitric oxide synthase activities and moisture content of spinal cord tissue at various peroid after spind cord injury, using technique of semi-quantitive reverse transcriptase-polymerase chain reaction(RT-PCR) and biochemistry. In the meantime, the histology was also examined. Results: 1 .The levels of bFGF mRNA increased significantly after spinal cord injury as compared With those in sham operation group (p<0.05). 2.The aboved indexes in bFGF group were lower than those of injuried group in different extent (p<0.05). 3.The histological examination showed that spinal cord injury was slight in bFGF group. Conclusion: 1 .More bFGF was produced by spinal cord itself and this kind of protective mechanism might play an important role in its selfrepamng and regenaratmg after spinal cord injury. 2.Exogenous bFGF can reduce the spinal cord edema, inhibit nitric oxide synthase activities and the formation of nitric oxide excessively after spinal cord injury.In this study, it is suggested that the mechanism of spinal cord edema after spinal cord injury is related to level of nitric oxide.
