Objective: Urethroplasty is a procedure to treat hypospadias and urethral stenosis. Hypospadias is an urogenital anomaly with an incidence of 1/125 50 live male births. The ventral chordee and meatal problem cause the cosmetic and functional abnormity. The complication includes fistula, urethral stenosis et al and it makes urethral reconstruction one of the most challenging procedures. The results of clinical and lab studies of the materials used in the procedures were reported and were helpful for the modifying of urethroplasty for the repair of hypospadias. Our study concentrates on the change of the inner face of prepuce graft after the operation . Methods: About 1cm penile urethral was mobilized and excised during the experiment. Inner face of the prepuce size 0. 8. 0 X 2. 0?. 5cm was harvested, and the tissues adhering to its back were eliminated. The graft was rolled around an 8F silicone tube, and its ends were anastomosed to the distal and proximal ends of the urethral respectively. At the 4?th day, 1st, 2nd, 3rd, 4th and 6th week after the operation the animals were sacrificed. During a second anesthetic induction the abdomen was opened and the aorta was isolated and cannulated with an 8F silicone catheter. 4Oml. India ink was injected. Then the specimen was harvested and the animal was sacrificed. Specimens were then fixed in formalin, stained using a standard HE protocol, and studied by light microscopy. Result: The grafts remained the characters of keratinized stratified squamous epithelium and the proximal urethral epithelium kept unchanged. Conclusion: Inner face of prepuce is an ideal graft for urethralplasty.
