Objectfr~:(1~fo establish a new type of cardiac xenotransplantation modeL (2) To evaluate the effect of complement depletion by using Chinese Venom Factor on hyperacute rejection in the cardiac xenoimnsplantation model. Methods:(1) Hear1~ obtained from guinea pig , was transplanted into abdomen of Wistar rat Before transplantation ,Ihe recipient was removed left kidney.The donor left common carotid artery was anastomosed to the recipient left renal artery by a 搒lecve?anastomosis. The 揷uffed? donor left pulmonary artery was inserted into the recipient left renal vein . (2)Some xenognifls from guinea pi~ were transplanted into abdomen of rat after the recipients being given i.p a dose of CVF 0.20 ii g.g?. Recipients were divided into A group (controlled), B group (only given CVF), C group (CVF + Cy + splenectomy) , D group (Cy +Splenectomy) .The histopatholo~r ,survival time of xenogmft,and level of serum AST~ CL CKMB were observecL Results: (1) 40 times of formal transplantation were done 1 with the successflul rate 95%.The mean time of operation was 63 minutes.(2) The survival time of xenog~aIt was from 15 minters to 52 hours. The histologic analysis showed that compared with the othei~ the groups given CVF had significantly difference .There were difference in A~ D groups which were contrasted to before and after transplantation of the level ASL CK~ CKMB, meanwhile a remarkable change was discovered among the phrase of survival time in B C groups~especialIy in 8h after transplantatiorL Condusions:(1) The method is easy , quick and reliable. This is a ideal model to study rejection of xenotransplantation, and to choose the new inimuno -suppressive agents.(2X2VF can prolong xenograft survival time by depleted complement activity and restricted hyperacute reject in this model.
