Objective: Measure the variation of complement components level before and after CVF used in different porcine to baboon xenotransplantation models,which are use transgene pig organs and tissues as transplant grafts.Investigate the potent of complement depression with the usage of hCRP expressed transgene graft combined with CVF therapy.Provide experimental gist for combined applications in pig-baboon xenotransplantation.Materials and methods:(1)Build up pig to baboon artery patch and kidney xenotransplantation models with GTKO / CD46/ CIITA and GTKO / CD46 transgene porcine artery patches and GTKO / CD46 /CD55 / TBM / EPCR / CD39,GTKO / CD46 / CD55 / EPCR / TFPI /CD47 and GTKO / CD46 / TBM transgene porcine kidneys provided by transgene pigs.(2)Give 100 U CVF to porcine - baboon kidney xenotransplantation models in-1 day and 0 day.(3)Collect baboons`blood samples at different time points and isolate the serum separately.(4)Measure the serum samples by C3 a,Bb,C1sC1 Elisa kits and analyze the data.Results:(1)Level of C3 a in samples took after the transplant surgery is13.33% and 21.04% increase compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group without the usage of CVF.(2)Level of C3 a in samples took after the transplant surgery is 87.24%,71.73% and 86.76% decrease compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group with the usage of CVF.(3)Level of C3 a in samples took after the transplant surgery is decrease about 10.23% and 10.22% in two patch xenotransplantation models but another patch xenotransplantation model is 8.58% increase compared to pre operation level in CD46 expressed transgene in artery patch xenotransplantation group without the usage of CVF.(4)Level of Bb in samples took after the transplant surgery is 2.45% and 63.93% decrease compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group without the usage of CVF.(5)Level of Bb in samples took after the transplant surgery is 198.84%,101.53% and13.41% increase compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group with the usage of CVF.(6)Level of Bb in samples took after the transplant surgery is decrease about 29.26% in one patch xenotransplantation models but another patch xenotransplantation model is 18.86% increase compared to pre operation level in CD46 expressed transgene in artery patch xenotransplantation group without the usage of CVF.(7)Level of C1 s insamples took after the transplant surgery is 41.50% and 63.12% decrease compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group without the usage of CVF.(8)Level of C1 s in samples took after the transplant surgery is 51.82%,53.89% and33.56% decrease compared to pre operation level in CD46 and CD55 expressed transgene kidney transplantation group with the usage of CVF.Conclusion:(1)The activation of complement system could not be depressed completely by using the gene modified pig`s tissue and organ individually.(2)Complement cascade is partly eliminated by the combination of CVF therapy and gene modified pig`s tissue and organ.
